Epidemiological Investigation Of Hepatitis E Virus On The Livestock And Fowls In The Western Yunnan Province

Objective1.Investigate and study the infections of hepatitis E virus in the serum of the swine in Dali in the western Yunnan province.Search the prevalence of hepatitis E virus in the livestock in Dali in the western Yunnan province.2.Investigate and study the infections of hepatitis E virus in the serum of the chickens and the migratory birds in the western Yunnan Province.Search the prevalence of avian hepatitis E virus in west Yunnan province,and proof the possibility of avian hepatitis E virus spread through the migration of migratory birds in the western Yunnan province.Methods1.273 serum samples of slaughter pigs were collected from the pig farm of Dali belong to the western Yunnan Province.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum HEV antibodies in slaughter pigs.Explore livestock epidemic situation of hepatitis E virus in the western Yunnan provice.2.1005 serum samples of chickens were collected from vegetable markets in the western Yunnan Province(Dali,Lijiang,Chuxiong,Baoshan,Ruili and Nujiang).Enzyme-linked immunosorbent assay(ELISA)was used to detect serum HEV antibodies in chickens.3.Migratory birds droppings collected from Dali Xiaoputuo and Heqing Caohai use RNA extraction Kits to extract total RNA.By AMW reverse transcriptase with R1 reverse transcription primers make total RNA into c DNA.Then use the RT-PCR method respectively by cDNA and the first round product as templates.After two rounds of amplification then use 1% agarose gel electrophoresis analysis,to test the existence of strips nearly at 359 bp and 221 bp.Results1.We collect and test total 273 slaughter pigs serum samples in Dali in the western Yunnan province from 2015 to 2016.The result shown that there are 5 serum anti-HEV Ig G positive acounting for 1.83%(5/273)and 3 serum anti-HEV Ig M positive acounting for 1.10%(3/273).Slaughter pigs hepatitis E virus infection rate in Dali in the western Yunnan Province is 2.93% in this survey.This survey of slaughter pigs hepatitis E virus positive rate compared with previous swine investigations' has significant differences(?2=25.85,p<0.05),and slaughter pigs hepatitis E virus positive rate of this survey is lower than the previous.2.We detect total 1005 sera samples of chickens from 2014 to 2016 in six cities belong to the Yunnan provice,and the highest area of avian hepatitis E virus infection rate in the western Yunnan Province is in Lijiang for 1.63%(3/184).Ruili,Baoshan and Nujiang are not detected in chickens antibodies against HEV.This survey finds that in the western Yunnan province the overall infection rates of chickens for hepatitis E virus is 0.9%(9/1005).3.We collect 183 fresh migratory birds droppings and test HEV RNA from 2015 to2016,but there are not amplified the desired strip that failed to proof the fact that migratory birds can carry hepatitis E virus.Conclusions1.The infection rates of swine hepatitis E virus infection rate is 2.93% in the western Yunnan province.2.Hepatitis E virus infection exists in the western Yunnan province,and hepatitis E virus has the infection rate for 0.90% in the western Yunnan province.3.In this survey,migratory birds in the western Yunnan province have not found the infections of hepatitis E virus.
