Primulina has many species and abundant colors,which flowering period is long and has potential as an ornamental plant.The study revealed the anthocyanin pigments in several plants of Primulina and Primulina swinglei flower and established the transgenic system of this genus would help for further studying the physiological and genetic mechanisms of the flower-color variation in Primulina,and is also helpful for creating new color germplasm by hybrid breeding or molecular breeding.The main findings are as follows:?1?The total content of anthocyanin in each species was measured by the pH difference method in genetic.The higher the total content of anthocyanin in the flower,the deeper color of the flower was found.Based on the different content,13 species were selected from the 38 species for the identification of anthocyanin components.Which the more color,the more anthocyanin.And Primulina swinglei had 17 kinds of anthocyanin,which was the most species of anthocyanin because of the different colors in petals and cylindrical fireworks.P.Mabaensis,P.huaijigenesis and P.Lutea had no anthocyanin,flavonoids as the main color material.?2?The petal present purple is mainly caused by the high proportion?up to 64.0%?of bluish violet pigment of delphinidin,malvidin and petunidin.The red cylinder is possibly caused by the high content of cyanidin and peonidin,which occupied 36.7% and 20.0%,respectively.The percentage and content of the pink pigment compounds increased and bluish violet pigment compounds decreased during flower development,probably due to the increment proportion of petal.Before the flower opening,the total content of pigment increased and then tended to be stable?there was a peak at stage S2?.After the flower opening,the pigment content decreased,probably due to the degradation of pigment.?3?In this paper,A rapid and efficient Agrobacterium tumefaciens-mediated transformation and regeneration system of Primulina was established.which depends on a powerful callus induction ability of leaf explants in P.huaijigenesis on MS medium containing 6 g·L-1 agar,30 g·L-1 sucrose,0.1 mg·L-1 TDZ and 0.1 mg·L-1 NAA.Here the most efficient shoot induction medium is MS medium containing 6 g·L-1 agar,30 g·L-1 sucrose,0.5 mg·L-1 TDZ and 0.1 mg·L-1 NAA;the most suitable initial antibiotic concentration of Cefotaxime was 500 mg·L-1 and later decline.After screening 4 times with Hygromycin(20 mg·L-1)to PCR identification,As outlined above,the conversion efficiency was about 20%.
