Lily has significant economic value,cultural value and medicinal value.In this paper,lilies located in Mountain Changbai Area are chosen as experimental materials for our research to carry out.Our results are showed as following:1.Lilium plants in Changbai Area have higher genetic diversities at the level of species,genetic diversity index of Shannon’s index(I)for L.cernuum is 0.4393,and for L.dauricum I is 0.4371、L.distichum I is 0.3975、L.pumilium I is 0.3948,L.lancifolium I is 0.3037.Results of genetic diversity research of populations:Genetic diversity of populations are significantly lower than level of species for L.distichum,the highest Shannon’s index is 0.2031,with great genetic differentiation and weak gene flow existing among populations(Gst=0.5248 Nm*=0.4527).The differentiation within population and among populations are extremely significant(P<0.0010),Mantel Test reflects there’s a significant positive correlation between genetic distance and horizontal geographic distance(r=0.0182,P<0.01),genetic drift is the main cause for variation of colonial genetic structure.Moreover Φ value is 0.581,means that selfed is their advantage mode of reproduction which accelerating the differentiation and weakening the gene exchange between populations of L.distichum.Genetic diversity of populations are lower than level of species for L.dauricum,the highest Shannon’s index is 0.3133,with great genetic differentiation among populations,and weaker gene flow but larger than 1 existing among populations(Gst=0.3053 Nm*=1.1379),gene flow could resist the variation of genetic structure causing by genetic drift.The differentiation within population and among populations are extremely significant(P<0.0010),Mantel Test reflects there’s no significant correlation between genetic distance and horizontal geographic distance(r=-.7002,P>0.05).Moreover Φ value is 0.417,means there’s hybridization and selfed their among populations,and this mode of reproduction weaken the differentiation of L.distichum populations.Because of the fewer natural resources of L.cernuum 、 L.pumilium 、L.lancifolium,just only one population,analysis among populations can’t be done.2.ITS2 primers are chosen as DNA barcode to finish the identification of L.pumilium.In the meantime,we find out 3 ISSR primers and then using them to distinguish five Lilium species located in Mountain Changbai area(UBC807、UBC826、UBC866).3.Analysis for the value of edible and medicinal: The protein content is low in L.distichum and L.dauricum of Mountain Changbai area(Are 0.99% and 0.44%,edible lily reference standard is 6.7%).The content of water soluble carbohydrate of these two species is higher(Are 69.65% and 76.18%,edible lily reference standard is 79.5%).The content cold malt extract is higher(Are 0.99% and 0.44%,Chinese pharmacopoeia criteria is ≥18%),then HPLC is used to analyze the medicinal ingredient of steroidal saponins and polysaccharide: there’s higher content of steroidal saponins in two lilies,and the content of steroidal saponins in L.distichum is significantly higher than L.dauricum,while a low content of polysaccharide for both.From the above,L.distichum and L.dauricum need to be protected,and L.pumilium 、 L.cernuum 、 L.lancifolium are all scarce resources in Mountain Changbai Area,protective measures must be adopted.In addition,both L.distichum and L.dauricum have certain medicinal developing value,with an obvious significance of protection for germplasm. |
