| Perinatal stage of dairy cows usually refers to prenatal three weeks to postpartum three weeks,also named as "transition",and the major inflammatory and metabolic diseases usually occur during the perinatal stage due to influence of various factors.Uterine infection is one of the primary causes of infertility in cows,which leads to the serious damage to the development of the dairy industry and serious economic losses.We tested relevant blood index of postpartum uterine infection in dairy cows,compared to the changes of physical and chemical index and related cytokines in blood of the healthy or sick cows,hoping to reveal the occurrence pattern of postpartum uterine infection.Thirty perinatal healthy cows have been chosen,and whole blood was collected from the tail vein to observe the changes of physical and chemical index and gene expression levels of related cytokines between prenatal 7 day and postpartum 30 day.Ten perinatal endometritic uterus-infected cows have been chosen,by using the same processing method to observe the changes of physical and chemical indexes and gene expression levels of related cytokines between the birth and postpartum 30d.1.Changes of physiological and chemical indexes of blood in perinatal healthy cowsWBC in prenatal 7d was at a higher level,increased significantly on the day of birth(p<0.01),and on postpartum 7d,the WBC levels decreased significantly to the levels that were lower than prenatal 7d(p<0.01).The result of Gran was in line with the variation trend of WBC.On the day of birth,Lymph and Mon continued to rise,without no significance(p>0.05);both Lymph%and Mon%decreased significantly on the day of birth(p<0.01);however,the Gran%increased significantly on the day of birth(p<0.01).RBC increased significantly on the day of birth(p<0.01),then began to fall gradually:RBC in postpartum 30d was significantly lower than that on prenatal 7d(p<0.01).Changes in HGB and HCT%were in line with the variation trend of RBC.PLT began to rise,the levels postpartum were significantly higher than prenatal levels(p<0.01).However,all variations were within the normal range.There was an obvious increase of AST,with a slight rise in TBIL,IDBIL,and ALT within the normal range,which indicated that liver cells were damaged,but did not cause liver function obstacle.CREA was at a high level during the childbirth,then dropped down to normal at 30 day postpartum.There was a little variation of BUN.2.Changes in gene expression levels of related cytokines of blood in perinatal healthy cowsGene expressions of IL-1α,IL-1β,IL-6 and TNF-a increased significantly on the day of birth(p<0.01).The levels of IL-8 increased significantly on both prenatal 7d and the day of birth,then gradually decreased(p<0.01).3.Changes of physiological and chemical indexes and related cytokines of blood in perinatal endometritic cowsWBC increased gradually after birth,and increased on postpartum 30d(p<0.05),which was significantly higher than the level in healthy cows(p<0.01).Gran%increased and was higher on postpartum 30d(p<0.01).However,Lymph%and Mon%continued to decline,decreased significantly on postpartum 30d(p<0.01).There was a significantly increase of AST,ALT,TBIL,and IDBIL in experimental group(p<0.01),CREA,GLU,Ca,and P in blood decreased after childbirth;TG was significantly higher(p<0.01).The gene expression levels of IL-1α,IL-1β,IL-6 and IL-8 were significantly higher than that of healthy cows(p<0.01),reached the highest level in postpartum 7d,and then decreased significantly in postpartum 30d.The gene expression level of TNF-a on the day of birth was lower than the level of healthy cows,then increased significantly in postpartum 7d,at last,decreased significantly in postpartum 30d,but remained higher than healthy cows(p<0.01).In conclusion,our results indicated that perinatal cow could cause the changes of the hematological parameters including white blood cells,red blood cells and blood platelets,but the changes of indicators were within the normal range.The WBC in endometritic cows was significantly higher than healthy cows,and the proportion of Gran%and Lymph%was seriously imbalanced.Accompanied with hepatosis,low GLU,and low Ca,the immune system was]ikely affected,which promoted the development of inflammation.The gene expression levels of IL-1α,IL-1β,IL-6,IL-8 and TNF-α had significant changes in both healthy and endometritic cows.However,the expression of these cytokines in endometritic cows were significantly higher than healthy cows,especially IL-6 and IL-8,indicating local and systemic inflammation and pathological damage when uterine infection happens. |
