Fritillaria taipaiensis P.Y.Li is one of the unique Chinese herbal medicines in Taibai Mountain Qinba Mountains.The dry bulbs of this plant are used as making lungs comfortable,eliminating sputum and relieving cough,which has always great concerns on the traditional chinese medicine.Fritillaria taipaiensis P.Y.Li sexual reproduction need 4-5 years to complete,with the seed low germination rate and germination difficulties.By asexual reproduction method,with a large amount,low propagation coefficient,high demand,the market has been in short supply,resulting in high production cost.The modern technology of tissue culture for rapid propagation of a large number of plant seedlings may be provided,at the same time,a flower can be strong 200-250 seeds,if the seed germination is also an important way to choose.Therefore,this study try to provide technical basis for large-scale artificial cultivation of Fritillaria taipaiensis P.Y.Li from three aspects,which including the tissue culture,seed germination and the impact of effects of light on the growth and development of plants.The results are as follows:1.The breeding method of tissue culture of Fritillaria taipaiensis P.Y.Li Tender stem segments was optimal for explant Primary culture.The best sterilizing method for tender stems was disinfected with 75% ethanol for 30 s,and then by 10% sodium hypochlorite for20 min,finally,soaking with 0.1% mercuric chloride for 5min.Washing 5 times with sterile water when the solution is replaced.The optimal medium for callus induction was MS+NAA1.0mg/L+6-BA3.0mg/L,which the rate of callus induction in this medium was the highest,and the growth rate was fastest.The optimal medium for regeneration induction of bulblet was MS+NAA1.0mg/L+6-BA0.5mg/L,which the rate of bulb induction was the highest.The optimal medium for regeneration induction of bulblet was MS+NAA0.5mg/L+6-BA0.5mg/L,which the rate of adventitious bud induction was the highest and the growth of adventitious bud was the most vigorous.2.The sand method of Fritillaria taipaiensis P.Y.Li seeds The best method of storing seed is as follows: First,dealing seeds with GA(50mg/L)for 24h;Second,threshing the seeds;Third,putting seeds in wooden box with the proportion of 1:1:1:2 for seeds,sawdust,humus and sand.And then,mixing periodic and covering with wet cloth to keep the humidity between 60% and 70%.Finally,keeping the room cool and wet.When storing 90 d,the embryo has reached 3/4 of the endosperm.At this time the activity of catalase and peroxidase increased dramatically,which suggested that the morPnology of the seed is finished basically after 90 days and can be entered into the low temperature storing.The activity of the two enzymes was slightly reduced when the temperature is low,and then had a sharp increase when the temperature is high.The Pnysiology of the embryo is ripe and can be sown after 210 days.3.The method of raising seedlings of Fritillaria taipaiensis P.Y.Li The sand and hormone treated seeds,selection of soil moisture,soil 1cm,seedling effect is best.4.Effect of illumination on the growth and development of Fritillaria taipaiensis P.Y.Li Shading rate is 60%,shading has the greatest effect on the growth of "leaves" period plant,which can promote its growth;followed by the "Tree son" period plant and the last one is "Lantern flower" period plant.Under the shading condition,the rate of Fritillaria taipaiensis P.Y.Li accomulation was fastest and the content was highest in "leaves" period plant.There was no significant difference in the effect of shading on the content of Fritillaria taipaiensis P.Y.Li in "Tree son" period plant.Shading is reducing the content of Fritillaria taipaiensis P.Y.Li in "Lantern flower" period plant.Therefore,it is recommended that only to shade the "leaves" period plant.
