Followed the guidance of traditional chinese medicine theory,using modern research means and production processes. Traditional Chinese detoxification medicine for HHD (Polysaccharides From Hedyotis diffsua Wild, PHDW) active ingredient extraction and purification,the ative ingredients in HHD (Polysaccharides From Hedyotis diffsua Willd, PHDW), which was Traditional Chinese medicine for detoxification, was extracted and refined in this study. Using the extraction process of PHDW by response surface analysis (RSA), Research temperature, ph and cellulase enzyme concentration three factors on the extraction rate of PHDW, Based on this design and response surface analysis and central composite optimization of the extraction process of PHDW. if the solid-liquid ratio is 1:10 extracting time is fixed at the time of 1h,the optimum extraction is that ph 7.8, temperature 55.7°C, the enzyme dosage of 2.5%. Under these conditions, the PHDW extraction rate of 9.83%. Use of the cellulase RSA analysis method to optimize Extraction of the PHDW. Get the Parameter of extraction conditions is accurate and reliable, cellulose extracted polysaccharides is High purity, Conditions easy to control, Can be used for large-scale production, Have a great practical value.80 healthy Kunming mice were randomly divided into normal control group (CN,given normal saline), immune suppression model group (Model,given normal saline) and PHDW low, medium and high dose treatment group (PHDW 50?100? 200mg/kg bw) a total of five groups. In addition to the CN group, the rest of each intraperitoneal injection CTX (100mg/kg bw) 3d; each gavage once daily continuously 15d, measured immune organ index, serum hemolysin,spleen lymphocyte proliferation and cytokines and other immune parameters. Result:Compared with control group, model group, spleen index, thymus index, serum hemolysin, NK cell activity, spleen lymphocyte proliferation and serum Il-2, 11-6, TNF-a levels decreased significantly (P<0.01 or P<0.05). Compared with M group, The middle and high PHDW doses can significantly improve the immune organ index mice hemolysin titer and half hemolysin, Polysassharides immunosuppressed mice splenocytes ConA and LPS-induced transformation a significant role in promoting (p<0.01), The middle and high PHDW dose range of mice immunosuppressive function of NK cell activity with a facilitating role(p<0.01). PHDW-L, PHDW-M, PHDW-H can significantly enhance the immunosuppressed mice serum 11-2,11-6, TNF-a content. Conclusion:Polysaccharides From Hedyotis diffsua Willd can enhance the immune function in mice.Used MTT assay observe mouse thymus and spleen lymphocyte metabolism and synergy with ConA and LPS-induced transformation for Polysassharides, Mouse Thymus and spleen macrophage metabolism and phagocytosis of macrophages and mouse thymocytes and splenocytes NK cell activity.Result:MTT assay on mouse spleen lymphocytes metabolism, collaborative ConA and LPS induced transformation has significant role in promoting for Polysassharides, When the concentration of 200ug/ml has the most obvious promoting, And the mouse thymocytes metabolism, collaborative ConA and LPS-induced no significant role in promoting transformation, it is no effect on mouse Thymus macrophage metabolism, phagocytosis; The mouse Thymocytes and splenocytes NK cell activity was significantly promoted, and showed a clear dose-effect two-way relationship and promote the most significant effect on the concentration of 150ug/ml.Conclusion:Polysassharides not only have significant role in promoting on mouse spleen lymphocytes metabolism and collaborative ConA and LPS induced transformation,But also macrophage metabolites and phagocytosis, the mouse thymus lymphocytes and macrophages obvious, NK cells Killing activity has a significant role in promoting in spleen and thymus.
