| Zhuanghe Dagu Chicken is an native breed with both excellent meat and eggs performance in China. It grows mainly in Zhuanghe county, Dalian city. It was identified as the geography trademark by the state administration for industry and on the books of National Livestock or Poultry Genetic Resource Protection List by Ministry of Agriculture in the age of 2006. The basic research is ignored at the same time of economic development. It has been past more than twenty years for the latest data collection and performance test. The genetic database has not been established related with its further promoted though many preserving seed bases work on the Protection of local fine varieties. This leads to a status with relatively rare of basic information in special genetic data. In this research, multiple microsatellite fluorescence PCR system has been established through twenty microsatellite marks and more than ninety specimens have been analyzed in order to explore genetic diversity, inbreeding degree and genetic transformation level for three preserving seed groups of Zhuanghe Dagu Chicken. The result will provide theoretical basis for Zhuanghe Dagu Chicken selection, production, exploitation and utilization.The results were as follows:1.Seven groups of multiple microsatellite fluorescence PCR system has been established. Four groups are dual-mode system, the other three groups are tripartite system. The test efficiency has been increased by double.2. Twenty microsatellite locis showed abundant polymorphism and total of 167 alleles were observed. Mean heterozygosities is 0.6111 and mean PIC is 0.6683. The average mean of Fis, Fst and Fit are 0.1431,0.0185 and 0.1269 respectively.The experimental results indicated that three populations of Zhuanghe Dagu chichen has abundant genetic diversity. Inbreeding and apparent genetic differentiation has not been exist. The results also indicated that over the years,the work of Dagu chicken breed conservation and selection were very successful. |
