Taking 13 Ganoderma lucidum strains as experimental material,which were identified,evaluated and genetic diversity determined by esterase isoenzyme zymogram analysis,ISSR molecular marker technique and fruiting morphology.The strain was screened by fruiting test and a wild strain was identified by ITS sequence analysis and domestication was carried out.The purpose of this study was to clarify the confusion of cultivar G.lucidum and the synonyms of the same species,and to provide excellent strains for production.The test results were as follows:(1)There were 12 bands with different relative mobility in the esterase isozyme spectrum of 13 strains of G.lucidum.The cluster analysis showed that the 13 strains could be divided into six groups when the genetic similarity level was about 80%,the first category included 2 strains,the second included 2 strains,the third included 2 strains,the fourth included 1 strain,the fifth included 5 strains,the sixth included 1 strain.(2)The results of ISSR cluster analysis showed that the genetic similarity level were about 0.74,the 13 strains were divided into 4 groups: the first group included two strains represented by "Gd1",the second group included 5 represented by "Gd3",the third included 2 strains of G.lucidum represented by "Gd9",and the fourth group included 4 strains represented by "Gd2".(3)The sequences of wild G.lucidum strains were amplified by rDNA-ITS and sequenced by Bioengineering(Shanghai)Co.,Ltd.The result of BLAST analysis indicated that the strain was G.lucidum.(4)The effects of six kinds of different stock culture mediums and five different kinds of mother spawn mediums on growth speed and condition of wild Inner Mongolia were studied.The results showed that the optimal stock culture medium were PDA + sawdust.The optimal mother spawn medium were 20% cotton seed shell,60% wood chips,18% wheat bran,1% sugar and 1% gypsum.(5)The biological characteristics of 13 Ganoderma strains such as biological efficiency,color and size of pileus,length and thickness of stipe were studied by production experiment.The results showed that the "Ganoderma amboinense" with high biological efficiency and the "Japanese Ganoderma lucidum" with large and thick pileus,high biological efficiency were suitable for cultivation in the midland and southern land of Hebei. |
