| Ganoderma lucidum is a traditional fungus with edible and medicinal value,which has been favored by scholars at home and abroad.Collection and identification of wild Ganoderma lucidum germplasm resources and analysis of active components are of great significance for rational development of wild Ganoderma lucidum in China,enrichment of germplasm resources in China and rational consumption of the public.In this study,30 wild strains collected from Nanzhao County,Henan Province were used as experimental materials,and their genetic relationship and germplasm resource identification were studied first.The fruiting bodies were obtained by domestication and cultivation,and their agronomic traits were determined.The polysaccharide,triterpene,soluble protein,crude protein and ash content of Ganoderma lucidum fruiting bodies were compared and analyzed.Finally,the antioxidant activity of Ganoderma lucidum water extract was determined:1.The results of esterase isozyme analysis showed that the Rf values of esterase isozyme spectrum of 30 wild Ganoderma lucidum strains were mainly between 0.11 and0.81,and 30 different bands were amplified.Through cluster analysis,the distribution range of similarity coefficients among 30 Ganoderma lucidum strains was 0.65 ~ 0.93.When the similarity coefficient was 0.709,30 wild Ganoderma lucidum strains could be divided into 7 categories,indicating that the polymorphism of the test samples was high.A total of 85 clear polymorphic fragments were amplified by ISSR molecular markers using seven selected primers.The size of DNA fragments was between 300 bp ~ 8000 bp.The genetic similarity of 30 Ganoderma strains was between 0.62 ~ 0.89.When the genetic coefficient was 0.728,30 strains could be divided into six categories.It was consistent with the esterase isozyme classification method.2.The results of ITS sequence analysis showed that strain YL-18 was Ganoderma ramosissimum.Strain YL-65 is Ganoderma tsugae.Strain NZ-27-1 is Clitopilus.The remaining 27 strains,such as YL-42 and YL-68,belonged to the genus Ganoderma lucidum.3.The results of domestication cultivation and agronomic traits analysis showed that30 strains could be divided into four types: fluffy,flocculent,branched and frost by mycelium morphology.The 30 strains were domesticated and cultivated,and 25 fruiting bodies of Ganoderma lucidum strains were obtained.The quality and quantitative traits were analyzed,and the domesticated Ganoderma lucidum strains were clustered.The results showed that 25 Ganoderma lucidum strains could be divided into five groups when the Euclidean distance was 3.92,and the strain YL-18 was self-contained(consistent with the results of esterase isozyme,ISSR molecular markers and ITS sequence identification).4.The results of active component analysis showed that there were significant differences in nutrient content between different strains of Ganoderma lucidum and the fruiting bodies of the same strain before and after domestication.The polysaccharide and ash contents of most wild Ganoderma lucidum were higher than those of their domesticated fruiting bodies.The triterpenoids,crude protein and soluble protein contents of some domesticated fruiting bodies were higher than those of wild Ganoderma lucidum.The antioxidant activity of water extracts before acclimation was higher than that of after acclimation,and YL-8,YL-68 and YL-40 showed better antioxidant capacity. |
