Up to the present,the breeding of C. militaris is still little known tothe world, mostly the wild strain was made Simple selection and domestication forcultivation or uses in producing directly. Aim to obtain fine strains of C. militaris,UVmutagenesis had been used.1. An optimum medium was determined, the compositions of fermentation were asfollows: Potato200g, glucose20g, KH2PO43g, MgSO43g, H2O1000mL.The activity ofpellets reached largest, and the fermentative period was6d. The optimal storage time ofliquid spawn in4℃was6d.2. Aim to obtain fine strains of C. militaris, fragments from the mycelia of C.militaris CM016, culturing in liquid medium, were diluted to104times by usingphysiological saline. Under a20-w UV lamp, the fragments were mutated for100seconds with a distance of32cm. Finally41C. militaris strains of which mycelia hadbetter morphological characteristics were obtained.3. Somatic incompatibility analysis indicated that15mutagenic C. militaris strainsof all had antagonistic lines with the original strain. Further isozyme analysis showedthat11of the15strains above had obvious differences with the original strain in geneticrelationships. Of the10mutagenic strains above,7ones were able to produce normalstromas.4.The strain mycelium growth condition, Agronomic character and fruit bodyyields were compared with8C. militaris strains, and correlation were analyzed in certainCharacters, The results showed that there existed the positive correlation relationshipbetween the mother cultures growth speed and culture spawn growth rate, no correlationbetween the production and mycelial growth rate and also no correlation between thefruit body yields and fruit body quality.C13grew fast in the culture with4.36mm/d. C7was the best strain in the liquid medium with the highest weight, the smallest diameter and the largest density. The experiment strains were selected by high temperaturetolerance primary. The result show that C6, C7and C16three strains were hightemperature tolerance. Compared with contaminated rate between regenerative strainsand original strains, C7and C16have stronger ability of anti-Trichoderma Viridecapacity. They also had a significantly higher production than the original strain. C3andC6had a significantly higher content of cordycepin than the original strain respectively.5. Two strains were obtained. The mycelium of C7grew better and faster, and hadstronger ability of anti-Trichoderma and Viride capacity. It also had a significantly higherproduction and it was the best strain in the liquid medium as a liquid strains.C6had highertemperature tolerance and higher content of cordycepin than the original strain.
