Bencaoruyanqing Pulvis is a natural medicine compound prepared by Inner Mongolia Huatian Pharmaceutical Co.,Ltd.With detoxification,eliminate carbuncle,swelling and pain,detoxification effect of poisonous.Objective: Through the identification of dairy cow mastitis pathogens,inflammation in vitro antibacterial,anti-inflammatory,acute toxicity and clinical treatment test,evaluated the curative effect of Bencaoruyanqing Pulvis,disinfected for dairy cow mastitis and its toxicity to the body,to provide scientific basis for the clinical application of Bencaoruyanqing Pulvis.Methods:(1)Mouse auricle swelling model,subcutaneous cotton ball inflammatory granuloma hyperplasia model and intraperitoneal blood vessels under the stimulation of acetic acid under the permeability model were used to observe the elimination of inflammation of Bencaoruyanqing Pulvis.(2)Staining,microscopic examination and biochemical experiments were used to identify pathogens in milk samples by Bacterial isolation and purification.(3)Oxford cup method and tube dilution method were used to determine inhibition zone,minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC).(4)Determination of the maximum tolerance dose(MTD)and maximum tolerance multiple of mouse were used to evaluate the safety of Bencaoruyanqing Pulvis.(5)Bencaoruyanqing Pulvis was feed by intragastric administration,clinical cure rate,and effective rate to evaluate the clinical effect of Bencaoruyanqing Pulvis to dairy cow mastitis.Results:(1)Anti-inflammatory test:the average swelling rate in high-dose group and medium-dose group of Bencaoruyanqing Pulvis were 32.49% and 37.01%,lower than that of saline control group,and the difference was extremely significant(P < 0.01);the average swelling rate in low-dose group was 45.62%,lower than that of saline control group,and the difference was significant(P < 0.05).The average granulation rate in high-dose group,medium-dose group and low-dose group of Bencaoruyanqing Pulvis were 52.85%,56.47% and 63.71%,lower than that of saline control group,and the difference was extremely significant(P<0.01).The average peritoneal fluid OD value in high-dose group and medium-dose group of Bencaoruyanqing Pulvis were 0.23 and 0.25,lower than that of saline control group,and the difference was extremely significant(P<0.01);the average peritoneal fluid OD value in low-dose group was 0.27,lower than that of saline control group,and the difference was significant(P<0.05).(2)Bacterial isolation and identification:141 strains of bacteria were obtained,4 species: Streptococcus dysgalactiae,Staphylococcus aureus,Escherichia coli and Staphylococcus epidermidis.The detection rates were 46.10%,24.11%,20.57% and 9.22%.(3)Antibacterial test:the MIC of Bencaoruyanqing Pulvis against staphylococcus aureus,escherichia coli and streptococcus mutans were 0.0625,0.125,0.0625g·mL-1;the MBC are 0.125,0.250,0.125g·mL-1.Bencaoruyanqing Pulvis was extremely sensitive against staphylococcus aureus,escherichia coli and streptococcus mutans,the diameters of the inhibition zone were 22.83,21.60,24.51 mm,all more than 20 mm.(4)Acute toxicity test:the maximum tolerable dose(MTD)of Bencaoruyanqing Pulvis was 600 mg / 10 g BW.The maximum tolerance multiple was 150 times,belong to the actual non-toxic,high security.(5)Clinical treatment test:Bencaoruyanqing Pulvis high dose and medium dose group treatment efficiency were 100%,the efficacy of the low dose group was slightly higher than that of the control group.Conclusion:Bencaoruyanqing Pulvis had a good effect on inflammation,having a significant inhibitory effect to main pathogens of dairy cow mastitis.Bencaoruyanqing Pulvis had good safety in normal dose of using and it could effectively treat dairy cow mastitis.
