The Ying Huang pulvis is belong to chinese herbal compound preparation,and its components include dandelion,Scutellaria baicalensis,fructus forsythiae and angelica.The prescription was improved over the traditional Chinese recipe.It is not only used to treat clinical sore or swollen with pus,give play to the effect of clearing away heat and toxic materials,but also have the ability to improve function or nourish one's vitality and blood.The prescription was used in the treatment of subclinical mastitis cows.This experiment is mainly by the methods of modern veterinary medicine to study the effect of anti-microbial in vivo,tanti-inflammatory in vivo,the effect of acute toxicity in mice and the effect of cure the recessive mastitis cows on the decoction of Ying Huang Pulvis.1.Isolation and identification of the pathogens from the subclinical mastitis Objective: Determine the species of the pathogenic microorganism from the sick cow,in order to make the drug's bacteriostatic ability targeted the pathogenic microorganism.Methods: collected 45 milk samples from the dairy cows with subclinical mastitis,determined the species of the pathogenic microorganism through germiculture,plate streaking and biochemical identification.Results: There were 3 kinds of pathogenic microorganism,respectively,staphylococcus aureus,Escherichia coli,Streptococcus agalactiae.Conclusion: The result was in accord with the common pathogenic bacteria in clinical outcome.2.The bacteriostasis experiment of Ying Huang Pulvis decoction to the pathogenic bacteria in vitro.Objective: To observe the antibacterial ability of Ying Huang Pulvis decoction to the pathogenic bacteria.Methods: To detect the drug susceptibility of pathogenic bacteria to Ying Huang Pulvis decoction through Oxford cup method;determined the minimum bacteriostatic concentration and minimum bactericidal concentration through constant broth dilution method.Results:The Streptococcus agalactiae,Staphylococcus aureus and Escherichia coli were sensitive to Ying Huang Pulvis(YHP)decoction,and the degree are highly Sensitivity,medium sensitivity and slightly sensitive respectively;The MIC of YHP to Streptococcus agalactiae,Staphylococcus aureus and Escherichia coli were 31.25 mg/m L,62.5 mg/m L,125 mg/m L.The MBC of YHP to Streptococcus agalactiae,Staphylococcus aureus and Escherichia coli were 62.5 mg/m L,125 mg/m L,250 mg/m L.Conclusion: The Ying Huang Pulvis decoction has a certain antibacterial effect in vitro.3.The anti-inflammatory effect of Ying Huang Pulvis decoction on inflammatory model of mice.Objective:To detect the anti-inflammatory effect of Ying Huang Pulvis decoction on the inflammatory model of mice.Methods: Mice were randomly assigned into five groups:Ying Huang Pulvis of the high-dose group(YHP-H),middle-dose group(YHP-M),low-dose group(YHP-L),the blank control group and aspirin group.Test of dimethylbenzene-induced mouse auricular tumefaction,acetic acid-induced changes in peritoneal capillary permeability and cotton pellet-induced granuloma were adopted to observe the anti-inflammatory effects.Evaluated the anti-inflammatory effect of Ying Huang Pulvis decoction through the absorbance of peritoneal fluid,the degree of auricle swelling of mice and the quality of granuloma;After continuous dosing,determined the spleen and thymus coefficient of mice and observed the influence of Ying Huang Pulvis on immune organs.Results: compared with the blank control group,the YHS-H can extremely significantly reduce the mice auricle swelling(P<0.01),YHP-M can significantly reduce the auricle swelling(P<0.05);Compared with the blank control group,YHP-H and YHP-M can significantly reduce the absorbance value of peritoneal fluid(P<0.05);The weight of subcutaneous granuloma caused by cotton in YHP-L and YHP-M were significantly lower than that in the blank control group(P<0.05);After 15 days' continuous dosing,the coefficient of immune organs in mice increased significantly(P<0.01)?Conclusion: The Ying Huang Pulvis decoction has a good anti-inflammatory effect and can significantly increased the coefficient of immune organs.4.Acute toxicity test of Ying Huang Pulvis decoction in mice Objective: To study the acute toxicity of Ying Huang Pulvis decoction in mice.Methods: Filled the mice with Ying Huang Pulvis decoction,and determine the maximum dosage in mice on the basis of preliminary experiment.Results: The result of preliminary experimental showed that the dosage at the maximum concentration and volume could not cause death in mice,then determined the Maximum administration dosage.When the maximum dosage was 140 g/kg,there was no death in mice.Conclusion: Ying Huang Pulvis decoction was non-toxicity medicine.5.The clinical trials of Ying Huang Pulvis on subclinical mastitis.Objective: To explore the therapeutic efficacy of Ying Huang Pulvis on subclinical mastitis.Methods: Give medicine to the dairy cow with subclinical mastitis at high,medium,low dosage group and establish a control group.After giving medicine,observed the development of their pathogenetic condition.Results:After treatment,the efficiently rate of Ying Huang Pulvis on subclinical mastitis are 95.83%?95.65%?84.00% respectively,the amount of somatic cell in milk was significantly decreased.Conclusion:The curative effect of Ying Huang Pulvis on subclinical mastitis was good.
