The single Chinese herbs and compound traditional Chinese medicines, which have the effect of killing and inhibiting adhesion on Mycoplasma suis (M.suis) in vitro, were screened in this paper. These findings can provide evidence for the clinical prevention and treatment of M.suis infection. The positive (containing M.suis) and negative (M.suis-free) blood were detected by direct microscopy and direct blood PCR, and positive red blood cells, negative red blood cells and M.suis pathogen were prepared. It was tested with qPCR under an in vitro maintain culture system for the change of M.suis copy-number in culture system at different culture times, the effect of M.suis copy-number in culture system under different medicines, the effect of M.suis copy-number on erythrocytes under different medicines, the adhesion effect of M.suis to erythrocytes after the interaction of different medicines with M.suis or erythrocytes, and the adhesion effect of M.suis to erythrocytes after the interaction of different concentrations medicines with M.suis or erythrocytes.The results showed that:(1) The copy-number of M.suis had not change significantly during the cultivated period.(2) In total 8 kinds of medicines, the killing effect of salvia miltiorrhiza, cortex fraxini and licorice was better with the killing ratio of 33.34%,30.21% and 30.15%, respectively. The killing ratio of artemisia annua, areca catechu, scutellaria baicalensis and sophorae was 25.92%,24.48%,22.58% and 22.14%, respectively. Cinnamon bark had the worst killing effect, which was 0.70%. The killing ratio of the compound Chinese medicines ?, ?, ? was 48.95%,39.50% and 41.04%, respectively.(3) The inhibiting adhesion effect of licorice was better, with the ratio of 62.18%. The inhibiting adhesion ratio of Salvia miltiorrhiza, artemisia annua, areca catechu, sophorae, cortex fraxini and scutellaria baicalensis was 34.04%,33.09%,26.19%,25.54%,24.70%, 22.77%, respectively, but ratio of cinnamon bark was 0. The inhibiting adhesion ratio of compound ? ?, ? was 82.20%,80.38%,71.79%,respectively.(4) The M.suis copy-number of adhesion to erythrocytes was reduced after the interaction of artemisia annua, scutellaria baicalensis, areca catechu, licorice, Cortex fraxini, sophorae, salvia miltiorrhiza, the compound Chinese medicine ?,?,?and doxycycline with M.suis. The M.suis copy number of adhesion to erythrocyte was reduced after the interaction of artemisia annua, scutellaria baicalensis, areca catechu, licorice, the compound Chinese medicine ?and ? with erythrocyte. The M.suis copy number on or to erythrocytes was not change significantly after the interaction of Cinnamon bark with M.suis or erythrocytes.(5) The M.suis copy-number of adhesion to erythrocytes was reduced after the interaction of the different concentrations of licorice, salvia miltiorrhiza, artemisia annua, cortex fraxini, the compound Chinese medicine ?,?, ? and doxycycline with M.suis. With the increase of medicine concentration, the M.suis copy-number on erythrocytes was reduced gradually. The M.suis copy-number of adhesion to erythrocytes was reduced after the interaction of the different concentrations of licorice, artemisia annua, the compound Chinese medicine ? and ? with erythrocytes. With the increase of medicine concentration, the M.suis copy-number of adhesion to erythrocytes was reduced gradually. The optimum concentration of cortex fraxin was 3000 ?g/mL, and other three kinds of single Chinese herbs, three compound Chinese medicines and doxycycline were 4000 ?g/mL.The results suggested that:(1) M.suis cannot proliferate in vitro culture system.(2) M.suis can be inhibited and killed by artemisia annua, scutellaria baicalensis, areca catechu, licorice, the compound Chinese medicine ? and ?, while the adhesive action of M.suis to erythrocytes was inhibited. M.suis could only inhibit and kill but not effect to erythrocytes by cortex fraxini, sophorae, salvia miltiorrhiza, the compound Chinese medicine III and doxycycline. Cinnamon bark could not effect on either M.suis or erythrocytes.(3) With the increase of medicine concentration, the effect of M.suis inhibiting and killing after the interaction of licorice, artemisia annua, salvia miltiorrhiza, cortex fraxini, the compound Chinese medicine ?, ? and ? was enhanced gradually. The ability of M.suis on erythrocytes after the interaction of licorice, artemisia annua, the compound Chinese medicine ? and ? was weakened gradually.
