Fumonisin B1(FB1) is the most prevalent member of fumonisins that are a group of mycotoxins produced primarily by Fusarium verticillioides and Fusarium proliferatum. In vivo studies have shown that FB1 induces pulmonary edema and liver toxicity, neurotoxicity and carcinogenicity, etc. The observed in vivo effects are similar compared with cell cultures exposed to fumonisins Experiments with several different cell lines have demonstrated that FB1 also induces apoptosis. The experimental research using human umbilical vein endothelial cells(HUVEC) for research materials, research FB1 vitro cytotoxicity HUVEC cells, which can supply laboratory evidences to evaluating the toxicity of FB1. The effects of FB1 on HUVEC cell proliferation, cell viability, cell cycle, oxidative stress, cell secretion, mitochondrial membrane potential, cell apoptosis and the expression of cell cycle- and apoptosis-related genes were detected by using the methods of fluorescence staining, flow cytometry, q RT-PCR, the results were as follows:(1) Methods HUVEC cells were treated with various concentrations(0,5,10,20?g/m L) of FB1 for different hours, and then design the blank media as a control group. Observation of cell morphology, cell viability measuring LDH levels, active oxygen content changes, analyze the toxic effects of FB1 on HUVEC cells. The results show that FB1 could significantly make cell morphology change, inhibit cell proliferation reduce cell viability, and induce oxidative stress injury in HUVEC cells.(2) Methods HUVEC cells were treated with various concentrations(0,5,10,20?g/m L) of FB1 for 48 hour.Observation the changes of HUVEC cells nuclear morphology, apoptosis rate, cycledistribution and mitochondrial membrane potential was futher investigated. result show that, FB1 induced HUVEC cells apoptosis Meanwhile,the apoptosis rate in each group was tconcentration-dependent. Cell cycle assay showed,FB1 mainly impacted on the G0/G1 phase in the cell cycle.(3) Different concentrations(0 and 10 ?g / m L) of FB1 treated HUVEC cells 48 h, measuring Caspase-3, Caspase-9, Bax and Bcl-2 and other related gene expression by RT-PCR method, the results show, Caspase-9 and Bax elevated levels of gene expression, Caspase-3 gene expression levels have increased to some extent, Bcl-2 gene expression levels have reduced, thereby inducing apoptosis in HUVEC.Conclutions: The toxic effects of Fumonisin B1(FB1) on HUVEC cells are indicated by inhibition of cell proliferation, cytokine Synthesis and Secretion, and cell apoptosis.
