Objective: To study the E.coli O157: H7 carrying status and pathogenicityof healthy cattle in Xinjiang beef main production area, the reporter isolates strains and establishes the animal illness model. At the same time Pulsatilla Decoction and Yujin San were used to cureand prevent the disease.Method: 1.Collecting 423 fresh cow dung samples from cattle farms in Yili district, Aletai district and Tacheng district of Xinjiang and after enrichment the reporter uses SMCA flat and MUG experiment to filter the bacteria. Then PCR technical and biochemical were used to identify the bacteria. Using PCR to detect the gene of Stx1, Stx2, eae A, tccp and Hly of E.coli O157: H7 in isolated bacteria. In order to determine the pathogenicity of the isolated strains, the reporter injects enrichment broth of EC bacteria strain to enterocoelia. 2.After grouping the kunming mice and injecting E.coli O157: H7 bacterium solution of different concentration, the strains of mouse LD50 were calculated by modified Karber's method, on the basis, the animal disease models were established. 3. In the Random allocation of healthy mice group, the reporter were injected with the bacterium solution according to the weight of mice. During the drug, the reporter were detected with the changing levels of the clinical symptoms, splenic coefficient and TNF-?, IL-17 in serum to determine the safety and the influence on immunization of Chinese Traditional Medicine formula. 4. The reporter were used Pulsatilla Decoction and Yujin San to prevent and cure the E. coli O157: H7 infected animal. From the changing levels of the clinical symptoms, splenic coefficient and TNF-?, IL-17 in serum to analysis the function of Chinese Traditional Medicine formula on animal immune system, bacteriostat and anti-inflammatory function.Results:1. There were one strain of E.coli O157: H7 in Yili district, Xinjiang and the relevance ratio is 1.176%. This strain carried all five detected poisonous genes(Stx1, Stx2, eae A, tccp, Hly). The relevanceratio in Aletai district and Tacheng district of Xinjiang was 0%. All the animals in experiments are died within 24 hours. 2. This experiment successfully established the animal disease model of E.coli O157: H7 infection, the strain LD50 was 1*105 cfu/m L. 3. There were no significant difference of spleen coefficient after the animals were irrigated with different concentrations of Pulsatilla Decoction and Radix Curcumae(P>0.05). 5d and 1d compared, the level of TNF-? decreased significantly(P<0.05)after irrigated with Yujin San. the content of TNF-? in other groups reduced extremely significantly(P<0.01). the high and low dose of Pulsatilla Decoction and the high and middle dose of Yujin San could significantly decrease the content of IL-17(P<0.05). the level of IL-17 in other dose of groups decreased extremely significantly(P<0.01). 4. The level of TNF-? and IL-17 in the serum of the animals infected with E.coli O157: H7 decreased to the before infection(P<0.05)after giving Pulsatilla Decoction and Yujin San.Conclusion: The cattle in Yili district carries the E.coli O157: H7 bacteria. From the results of animal challenge test, we can say that this bacteria has strong pathogenicity on human, and animal disease model were successfully established. The experiments showed that Pulsatilla Decoction and turmeric powder have nontoxic side effects, with two prescription treatment, it was found the routine dose of two recipes can decrease the E.coli O157: H7 infection on the effect of damage to the body, and these prescription have a strong antiinflammatory ability.
