Abstract/Summary:
Wolfberry(Lycium bararum L.)is Solanaceae and lycium L.plants. Which is a kind of medicinal and edible plant and has important medicinal value and economic value. Xinjiang as one of the main producing areas of wolfberry, has rich germplasm resources. Due to the long-term natural selection and artificial selection, genotype is high heterozygosity and the genetic background is very complicated, the research on genetic variation of germplasm resources from the perspective of molecular biology is often lagging behind the practice of breeding, which bring trouble to the effective utilization, the didentification of germplasm resources and the selection of parents, greatly affect the efficiency of genetic breeding. Therefore, study on genetic diversity of wolfberry germplasm resources in Xinjiang, which is conducive to the germplasm resources evaluation, breeding and rational utilization of new varieties. In this experiment, a total of 30 samples of cultivars(lines) and wild species in Xinjiang wolfberry germplasm resources garden were used as materials, using SRAP and SCoT markers to analyze the genetic diversity of 30 wolfberry germplasm resources, and DNA fingerprinting database was constructed. Jujube taxonomy is belong to Rhamnaceae and Ziziphus Mill.plants, it is a special fruit tree native in China, not only has economic benefit but also which is ecological economic forest tree species, due to its unique geographical location and unique climate conditions, Xinjiang has a rich variety of resources, the southern part of Xinjiang has become one of the main cultivation areas of jujubes. this experiment is based on the previous research, the jujube germplasm resources of different cultivation types in Xinjiang main producing areas were used as materials, SCoT markers were used to analyze the genetic diversity of the germplasm resources, and the DNA fingerprint database way also established, the aim was to provide the theoretical basis for the protection, identification,exploitation and utilization and breeding practice of jujube germplasm resources in Xinjiang. The main results are as follows:1. SRAP markers were used to study the genetic diversity and phylogenetic relationship of genetic resources of wolfber(Lycium bararum L.)in Xinjiang. The results are:(1)A total of 310 loci were produced by 12 pairs of SRAP primers, and polymorphic loci were 264 which accounted for 84.61% in the total amplified fragments.The polymorphism information content(PIC)values of these markers varied from 0.76 to 0.93 with an average of 0.83. Mean values of observed number of alleles(Na),effective number of alleles(Ne),Nei’s gene diversity(H)and Shannon’s Information index(I) were1.8461,1.3869,0.2280 and 0.3520.(2)Range of genetic similarity(GS) was 0.590 3 to 0.903 2 among 30 wolfberry samples, cluster analysis showed that samples could be divided into 2 groups and 5 subgroups accordingly at the GS of 0.70 and 0.73.The principal coordinate analysis and clustering results are basically consistent. Research shows, The genetic diversity of germplasm resources of wolfber in Xinjiang was relatively abundant, the genetic difference between wild species and Cultivars(lines) was relatively large,show a distant relationship, and the genetic difference among cultivars(lines) was relatively small, show a close relationship.2. The purpose of this experiment is to construct a DNA fingerprinting database of 30 wolfberry germplasm resources in Xinjiang,and the genetic diversity was analyzed based on start condon targeted polymorphism(SCoT), The results are: Among the 30 wolfberry samples,9 SCoT primers produced 256 bands,of which 219 were polymorphic bands,and the percentage of polymorphic rate was up to 85.62%,The polymorphism information content(PIC) values of these markers varied from 0.77 to 0.91 with an average of 0.85.The average value of the observed number of alleles(Na),effective number of alleles(Ne),Nei’s gene diversity(H)and Shannon’s Information index(I) were1.8562,1.4350,0.2611 and 0.3989.Range of genetic similarity(GS) was 0.5938 to 0.8398 among 30 wolfberry samples,cluster analysis showed that samples could be divided into 2 groups and 4 subgroups accordingly at the GS of 0.65 and 0.72.The principal coordinate analysis and clustering results are basically consistent, In addition, the DNA fingerprints of 30 wolfberry samples was constructed by using the 5 SCoT primers.The wolfberry germplasms in Xinjiang have a high level of genetic diversity,SCoT molecular markers are suitable for the analysis of genetic diversity and constructionof DNA fingerprinting of the germplasm resources.Moreover,It provides a wealth of genetic basis for evaluation, identification and selection of new varieties of the germplasm resources.3. SRAP and SCoT molecular markers were used to detect the genetic diversity of genetic resources of wolfber(Lycium bararum L.)in Xinjiang. The results are: Genetic diversity parameters and the average genetic similarity values(GS) from SCoT markers were all higher than those from SRAP markers, indicated that the SCoT marker has a greater marker efficiency and a higher genetic diversity detection capability,SRAP+SCoT markers PPB, PIC, Na, Ne, H, I and other genetic diversity parameter values ranged between two markers, higher than SRAP markers, cluster analysis based on combination of two molecular markers data showed that the range of GS was 0.61 to 0.84 among 30 wolfberry germplasm with a high genetic diversity, and all resources could be divided into 2 groups and 4 subgroups accordingly at the GS of 0.69 and 0.74.The principal coordinate analysis and clustering results are basically consistent.SRAP+SCoT markers can be more comprehensive to reveal the genetic diversity among 30 wolfberry germplasm resources in Xinjiang, Combined use can be beneficial to reduce the deviation which cause from the study on genetic diversity of germplasm resources of wolfberry by single molecule marker.4.The purpose of this experiment is to construct a DNA fingerprinting database of 42 jujubes Germplasm Resources planted in Xinjiang,and the genetic diversity was analyzed based on start condon targeted polymorphism(SCoT).The results are:326 bands were amplified by 9 SCoT primers,and thepercentage of polymorphic rate was up to 94.34%,The polymorphism information content(PIC) values of these markers varied from 0.79 to 0.93 with an average of 0.86. The average value of the observed number of alleles(Na), effective number of alleles(Ne),Nei’s gene diversity(H)and Shannon’s Information index(I)were1.9434, 1.3866, 0.2473 and 0.3905. The average GS value of genetic similarity coefficient was 0.7259,cluster analysis showed that samples could be divided into 4 groups and 4 subgroups accordingly at the GS of 0.65 and 0.67. The principal coordinate analysis and Clustering results are basically consistent, In addition, the DNA fingerprints of 42 jujubes samples was constructed by using the 9 SCoT primers. The jujubes germplasms in Xinjiang have a high level of genetic diversity, SCoT molecular markers are suitable for the analysis of genetic diversity and construction of DNA fingerprinting of the germplasm resources.Moreover, It provides a molecular biological foundation for evaluation, identification and selection of new varieties of the germplasm resources.
