Anions exist widely in the living body and the natural environment, and have a tremendous impact on our live.Among them, chloride ions, fluoride ions, and phosphate ions play an important role in life activities.Their level of content is closely related to physiological and pathological conditions.Fluorescent probes have the advantages of high sensitivity, low detection limit and convenient application, and they are widely used. Therefore,it is of great scientific significance to properly construct fluorescent probes capable of specifically detecting anions. In this paper, three molecular fluorescent probes were synthesized using simple reagents for the detection of chloride, fluoride, and phosphate anions, respectively. The specific research content is divided into the following three tasks:A chloride fluorescence probe MQ-TPP was designed and synthesized using 6-methoxyquinoline as fluorophore.MQ-TPP has good water solubility, and response time to chloride ions is less than 60 seconds. The probe has good selectivity and sensitivity to chloride ions with a detection limit of 130.3 μM.MQ-TPP was used to detect the chloride ion content in the serum.2.Using a commercial fluorescent dye resorufin as fluorophore and t-butyldimethylchlorosilane as the recognition group,a fluorescence sensor S-F for the specific identification of fluoride ion was designed and synthesized.S-F has good selectivity for fluoride ion,and the detection limit is as low as 84.3 nM.The S-F has a high sensitivity, and the 30 μM fluoride ion can increase the fluorescence emission by about 40 times.S-F responds to fluoride ions under weak alkaline conditions, the color changes significantly, and can be used to detect fluoride ions in the naked eye.The cytotoxicity of S-F is very low,and we have successfully applied it to the detection of fluoride content in toothpaste and imaging of exogenous fluoride ions in HepG2 cells..3.Resorufin was used as the fluorophore and methyl oxalyl as the recognition group.A reactive fluorescent probe S-pi for detecting phosphate anions was successfully synthesized. The probe has good selectivity for phosphate anions and can distinguish between phosphate anions and pyrophosphate anions. After responding to the phosphate anion under weak alkaline conditions, the color of the probe changes significantly and can be used to detect phosphate anions with the naked eye.The linear range of probe detection is 0.1-5.0 mmol, and 5.0 mmol of phosphate anion can cause a 50-fold increase in fluorescence emission.S-pi has good biocompatibility and has been successfully applied to HepG2 cell imaging.
