Peroxynitrite is a kind of active nitrogen.It is a very strong biological oxidizing agent with high reactive activity and strong nucleophilic activity.Peroxynitrite can cause a series of human diseases,including acute chronic inflammation,ischemic attack,diabetes and so on.Because peroxynitrite is closely related to life activities,so a highly sensitive tool is needed to detect it.Superoxide anion is very important for human body.As a normal metabolite in cells,it plays an important role in many physiological and pathological processes of life.Therefore,it has important research significance and application value to develop and design a fluorescent probe which can quickly detect the change of superoxide anion content.In a word,this paper has designed a benzonitrile-based fluorescent probe for detection of peroxynitrite and superoxide anion.The main contents are as follows:1.With benzonitrile as a fluorophore,two fluorescent probes,T1 and T2,were designed and synthesized for detection of peroxynitrite.The probe T2 is highly selective for peroxynitrite and it is a ratiometric fluorescence probe.It has good sensitivity for detecting peroxynitrite,and the detection limit is 1.8 n M.Furthermore,the probe T2 was successfully applied to the fluorescence imaging study for detection of the exogenous peroxynitrite in the Caenorhabditis elegans.2.With benzonitrile as a fluorophore,a fluorescent probe T3 for detecting superoxide anion were designed and synthesized.The probe T3 has very high selectivity and fast response for superoxide anion,the detection limit is 35.9 ? M.Therefore,the probe T3 is a good fluorescence enhancement fluorescent probe for detecting superoxide anion.
