Oyster polysaccharides are one of the potential materials for health food and drug.Numerous studies of preparation technology for oyster polysaccharides were processed.For efficient preparation of oyster polysaccharides,the ultrasonic extraction of oyster polysaccharides needs to be studied.Thus optimal ultrasonic extraction of oyster polysaccharides will be obtained.And then the anti-aging mechanism of oyster polysaccharides was studied.The results will be helpful for the application of oyster polysaccharides.Specific studies are as follows:1.The optimal ultrasonic extraction conditions of oyster polysaccharides were studied by a single factor experiment and an orthogonal test.The results showed that the optimal conditions of ultrasonic extraction were solid-liquid ratio of 1:15,ultrasonic power of 190 w,ultrasonic processing time of 35 min.The yield was 29.5% under optimized conditions.2.The experiment indicates that treatment with CGPS-1(400 mg/kg BW)improved the D-Gal-induced mice eating with model group.Three different doses of CGPS-1 no significantly affected the liver index,brain index,kidney index,thymus index and spleen index in D-Gal-induced mice.Treatment with CGPS-1(100 and 200 mg/kg BW)significantly increased the activities of SOD in serum of D-Gal-induced mice.Treatment with CGPS-1(100,200 and 400 mg/kg BW)significantly decreased malondialdehyde(MDA)content in serum of D-Gal-induced mice.3.Quantitative proteomic analyzed the effects of treatment with CGPS-1 on serum of D-Gal-induced mice by label-free LC-MS.The treatment with CGPS-1 on serum of D-Gal-induced mice analyzed using LC-MS/MS after removing the high abundance ratios protein.The Maxquant software(ver1.3.0.5)were used to search bioinformatics based on label-free LFQ data.The 4624 peptides and 541 proteins were identified by label-free LFQ data.The 20 differentially expressed proteins were analyzed by t-test.Among the 20 differentially expressed proteins,one protein no found in mice protein sequence databases.The 19 differentially expressed proteins covered 15 biological processes.They covered 8 molecular functions.They also covered 5 cellular components.Among these differentially expressed proteins,the three differentially expressed proteins as alpha-1-antitrypsin 1-5(AC:Q00898)(up-regulated),complement component 4B(Childo blood group)(AC:B2RW7)(up-regulated)and adenylyl cyclase-associated protein 1(AC:P40124)(down-regulated)have regulation of enzyme activity.The five differentially expressed proteins as carbonic anhydrase 1(AC:p13634)(up-regulated),peroxiredoxin-2(AC:Q61171)(up-regulated),malate dehydrogenase(AC:P14152)(up-regulated),xanthine dehydrogenase/oxidase(AC:Q00519)(down-regulated)and serum paraoxonase/arylesterase 1(AC:P52430)(down-regulated)have catalytic activity.These data demonstrated that CGPS-1 possess anti-aging effect in D-Gal-induced mice by enhancing the activities of scavenging free radical.These results suggests CGPS-1 can be developed as a new anti-aging health food and drug.
