Oyster polysaccharides have obvious anticoagulant,hypolipidemic,antithrombotic,prevention and treatment of cardiovascular diseases,improving immune function and anti-leukopenia and other biological activity,polysaccharides in oysters accounted for 20%-40%of dry weight,therefore,oyster polysaccharide became the focus.In this paper,the water-extract-alcohol-precipitation was combined with trypsin enzymatic extraction of oyster internal organs polysaccharides?CGOP?.Single factor experiments,the enzymatic hydrolysis time,enzyme dosage and water to feed ratio,were designed to investigate the effect on the purity of oyster gut polysaccharides.Based on the single factor experiments,three factors and three levels of orthogonal experiment were carried out with the time of water bath,the amount of trypsin added and the ratio of water to material.The optimum technological conditions for extracting visceral polysaccharides from oyster gut were determined.The results showed that the order of factors influencing the purity of CGOP was water bath time>water to material ratio>enzyme dosage,and the optimal combination was A3B2C2,i.e.5 h of water bath time,3:1 of water to material ratio,0.8%of enzyme dosage.The total sugar content of CGOP was 43.92%.The results of IR indicated that the extracted CGOP had typical polysaccharide IR spectra.Secondly,the anti-oxidative ability of CGOP was tested.The experimental results showed that CGOP showed some antioxidative effects on the four oxidation models.Twenty mg/mL CGOP could scavenge 95.7%DPPH radical,96.7%ABTS+radical and 99.4%hydroxyl radical,respectively.CGOP had better FRAP reducing ability.In CCl4-induced acute liver injury model,high dose CGOP?800 mg/kg·d?significantly reduced ALT and AST activity in serum,significantly reduced the content of MDA in liver tissue and increased the activity of SOD in liver tissue.Liver morphological observation,CGOP dose group can reduce the liver injury.This study shows that CGOP has a good antioxidant capacity,and can effectively protect the damaged liver tissue.The results provide the basic data for better utilization of oyster resources.Finally,according to"Pharmacopoeia of the People's Republic of China"?2015 edition?and other data,the evaluation criteria of oyster polysaccharide composite tablets were established from five aspects of the appearance of tablets,the difference of tablet weight,hardness,friability and disintegration time.The effects of microcrystalline cellulose,magnesium stearate and maltodextrin on the evaluation score of the tablets were studied.The orthogonal experiment of three factors and three levels was carried out based on the single factor experiment results.The results of orthogonal experiment showed that the order of factors affecting the evaluation score of tablets are as follows:magnesium stearate,microcrystalline cellulose and maltodextrin,the optimal combination is A2B2C3,namely microcrystalline cellulose,maltodextrin,hard Magnesium stearate dosage were 8%,2%,2%.Error D column was the great difference than the C column,which indicated that the levels selected by the level of the difference were not significant difference.
