| Oyster polysaccharide(CGPS-1)is one of important active substance from oyster,which was widely applied in food,health food and drug field.Numerous processes for oyster polysaccharide were to be study.Then the safety of CGPS-1 was to be study.In this study,the enzymatic extraction and drying process were studied.The preparation process of CGPS-1 was improved for extraction rate.The safety of CGPS-1 was evaluated by the acute oral toxicity and three terms of hereditary toxicity of mice.The results will be helpful for the application of CGPS-1.Specific studies are as follows:1.The optimal tryptic extraction conditions and of CGPS-1 were studied by a single factor experiment and an orthogonal test.The results showed that the optimal conditions of tryptic extraction were digestion temperature of 41℃,enzymatic time of 2 h and enzyme amount of 0.6%.The yield was 15.75% under optimized conditions.2.Polysaccharides were extracted from oyster using rotary evaporation-dried,spray-dired and free-dried treatments(REDCGPS,SDCGPS and FDCGPS).Among them SDCGPS has most reducing sugar。Value of reducing sugars in SDCGPS was high than REDCGPS and FDCGPS.All three polysaccharides showed typical polysaccharic absorption bands in FT-IR.Morphology of the dried three polysaccharides was analyzed by SEM.SDCGPS was observed dispersive small spherical particles.REDCGPS was observed larger flaky particles.FDCGPS was observed polyhedron shape with corner angles.Molecular mass of three polysaccharides was determined by HPLC.REDCGPS and FDCGPS have large molecular mass.SDCGPS has small molecular mass.The results showed that glucosidic bond of oyster polysaccharides was broke by spray-dired process,and then molecular mass of oyster polysaccharides changed small molecules with antioxidant activity.3.According GB15193.1-2014 National standards for food safety-acute oral toxicity test,No toxicity was found when acute oral toxicity dose was 20 g/kg BW in ICR mice(female and male,LD50>20 g/kg BW).Ames test showed that CGPS-1 has no mutagenic effect.The micronucleus calculation of mouse bone marrow cells showed negative results.Chromosome aberration test of CHL cells in vitro showed no significant abnormal splitting of chromosome.These results will be help for preparation process and application of CGPS-1. |
