Xinjiang Tamarix hohenackeri was selected as raw materials,the total flavonoids in the leaves was chosen as the research object,we studied on the extraction and purification process,compared and established three types of determination method,and some fundamental research on antioxidant activity and antibacterial activity of Tamarix hohenackeri flavonoids,it provides the experimental basis for the application of Tamarix hohenackeri flavonoids in the future the.The results are as follows:(1)The effects of different extraction methods on the yield of total flavonoids in the leaves of Tamarix hohenackeri was studied.Microwave ultrasonic assisted extraction(MUAE),microwave-assisted extraction(MAE),ultrasonic extraction(UE),accelerated solvent extraction(ASE)were used to extract the total flavonoids on the leaves of Tamarix hohenackeri,and the extraction method of ultrasonic microwave extraction and accelerated solvent extraction was optimized by response surface method.The results showed that the extraction yield of flavonoids by accelerated solvent extraction was the highest.The optimum technological parameters of the accelerated solvent extraction were as follows: 75% ethanol concentration,extraction temperature of 80 DEG C,status circle of 5 times and extraction time of 5min,under the optimum extraction condition,the yield was 40.78mg/g.The best process parameters of MUAE were: ethanol concentration of 61.56%,microwave power200 W,material liquid ratio 1:15,extraction temperature of 55 DEG C,extraction time was 3min,under this condition,the yield of flavonoids was 13.99mg/g.(2)Screened the optimal purification process of total flavonoids from leaves of Tamarix hohenackeri.D101 type resin was selected to purify the flavonoids of Tamarix hohenackeri leaves.The optimum purification conditions were sample concentration 0.2mg/mL,pH=1,flow rate of 1mL/min.The experimental results showed that the average purity was 75.04% when the purifying went under the best combination.(3)The study concluded the advantages and disadvantages of various methods through comparison and establish a method for determination of three kind of UV method and FS method for the determination of flavonoids in leaves of Tamarix hohenackeri leaves.UV visible spectrophotometry and molecular fluorescence spectrophotometry,the operation is simple and fast,but the molecular fluorescence spectrometry testing and debugging system stability is not good,to determine theoptimal conditions of the process is time-consuming.(4)By using a chemical method to determine the antioxidation activity of the flavonoids from the leaves of Tamarix hohenackeri,and compared with Vc.And the bacteriostatic experiment were discussed and the effects of temperature and pH value on the antibacterial effect was studies by the filter paper disk diffusion method.The experimental results show that the flavonoid has certain antioxidant activity.The antioxidant activity of flavonoids on the leaves of Tamarix hohenackeri was slightly lower than Vc,and within a certain range of concentrations,the antioxidant activity and the sample concentration is proportional.Flavonoids on the leaves of Tamarix hohenackeri have significant inhibitory effect on Bacillus subtilis,bacillus,Staphylococcus aureus,Escherichia coli and white slime,when the temperature is 70 DEG C,the medium pH was 5,the antimicrobial activity of flavonoids are thehighest.
