Berberine Chloride is an isoquinoline alkaloid commonly found in baberry and scutellaria.It has a variety of pharmacological functions in clinical,such as antibacterial,anti-cancer,anti-inflammatory,antivirus,treatment of cardiovascular disease,and so on.Because of its significant clinical efficacy,side effects,low toxicity,low price,therefore,berberine chloride has a wide range of market applications.At present,the synthesis of berberine chloride is widely concerned,there are shortcomings in the synthetic methods reported in the literature,and it is urgent to develop a new route with shorter route and higher efficiency.In this paper,two synthetic routes of berberine chloride were designed and optimized,and the optimum reaction yield was expected.All the intermediates were analyzed by 1HNMR,13CNMR,mass spectrometry,melting point and infrared confirm.Chapter 1 summarizes the pharmacological effects,structure-activity relationship,biosynthetic pathway and the synthesis of berberine hydrochloride.Chapter 2 synthesizes berberine chloride by linear synthesis strategy.Berberine chloride was synthesized starting from pyrocatechol in overall 14 % yield.The key steps include a Hofmann rearrangement,chloromethylation,nucleophilic substitution,hydrolysis,decarboxylation and cyclization.Chapter 3 uses the convergence strategy to design and synthesize berberine chloride.The convergent synthesis route is based on 1,2-methylenedioxybenzene as the starting material in overall 33 % yield.The key steps include include the Grignard reagent nucleophilic ring,deprotection and cyclization.In this paper,two synthetic routes are designed and synthesized,which are characterized by easy availability of raw materials,short route and high yield.They have the value of industrial application.
