| Berberine is an alkaloid.It is mostly found in plants of the genus coptis and berberine.In vitro,it has broad antibacterial activity against a variety of gram-negative bacteria and some fungi.In recent years,with the progress of interdisciplinary research in chemistry and medicine,this ingredient has shown superior pharmacological properties.Using the anti-inflammatory and regulating ability of berberine to respond to stress proteins as a way to prevent diabetic encephalopathy;the effect of inducing autophagy in hepatocytes mediates its function in inhibiting liver fat deposition,elucidating the mechanism of weight loss,etc.The relevant research results are the rediscovery of the effect of this drug,and also endow this kind of traditional medicine with brand new value and the meaning of continuing in-depth exploration.This paper studies the effect of berberine hydrochloride(BH)on the folding of protein amyloid fibrosis from the molecular level,and analyzes its reaction mechanism in combination with the interaction mechanism with serum albumin,in order to explore a variety of degenerative neurological diseases,such as Alzheimer’s disease.From the perspective of microorganisms,by analyzing the effect of berberine hydrochloride on the growth and metabolism of some drug-resistant bacteria,it is expected to further expand the antibacterial spectrum of the drug,and to explore its new capabilities against the drug resistance problem caused by the abuse of traditional antibiotics.Based on the rediscovery of drug action,this paper studies from two aspects:biological effect and antibacterial activity,and clarifies the interaction mechanism between berberine and serum albumin and its antibacterial effect on drug-resistant bacteria.Clinical application and other aspects provide certain scientific basis and theoretical support.The specific research work is as follows:1.Studies on the mechanism of interaction between berberine hydrochloride and serum albumins.The interaction mechanism between berberine hydrochloride and serum albumin was analyzed by UV-visible absorption spectroscopy,variable temperature fluorescence spectroscopy,three-dimensional fluorescence and synchronous fluorescence.By analyzing the data of variable temperature fluorescence and UV-visible absorption spectra,there are enthalpy-driven hydrophobic and hydrogen bonding forces between berberine hydrochloride and serum albumin,and the mechanism of action is static quenching.The effects of berberine hydrochloride on the secondary conformational changes of serum albumin were investigated by three-dimensional fluorescence and circular dichroism.2.Studies on the effect of berberine hydrochloride on serum albumin amyloid fibrosisUsing fluorescent probes,the effect of berberine hydrochloride on serum albumin was investigated by fluorescence spectrophotometry(FL),ultraviolet absorption spectroscopy(UV),circular dichroism(CD),field emission scanning electron microscopy(SEM)and other characterization methods.Effects of amyloid fibrosis.Data analysis showed that BH had inhibitory effect on amyloid fibrosis of BSA.Circular dichroism and SEM data also indicated that the addition of berberine hydrochloride could prevent the transition of protein secondary structure fromα-helix toβ-sheet.3.Studies on antibacterial activity of berberine hydrochloride against Saccharomyces cerevisiae,methicillin-resistant Staphylococcus aureus,multidrug-resistant Enterococcus faecium and multidrug-resistant Enterococcus faecalisThe experiment analyzed the metabolism of berberine hydrochloride on Saccharomyces cerevisiae,methicillin-resistant Staphylococcus aureus,multidrug-resistant Enterococcus faecium,and multidrug-resistant Enterococcus faecalis by microcalorimetry and optical density detection method.The antibacterial properties of the drug were evaluated by analyzing the growth curve and the effect on cell proliferation.The experimental results show that berberine hydrochloride has an inhibitory effect on the growth of yeast cells and three drug-resistant bacteria.The analysis of calorimetric data shows that berberine hydrochloride has an inhibitory effect on Saccharomyces cerevisiae,methicillin-resistant Staphylococcus aureus,and multi-resistant bacteria.The half inhibitory concentrations(IC50)of drug-resistant Enterococcus faecium and multidrug-resistant Enterococcus faecalis were 50.1μmol/L,68.8μmol/L,110.6μmol/L and 0.86μmol/L,respectively.As the reference group,the half inhibitory concentrations(IC50)of methicillin-resistant Staphylococcus aureus,multidrug-resistant Enterococcus faecium,and multidrug-resistant Enterococcus faecalis were 3.01μmol/L,0.86μmol/L,1.15μmol/L.Through the experimental results of the reference group,it was found that the bacteriostatic effect of BH on drug-resistant bacteria was more prominent.In this paper,the studies of the effect of berberine hydrochloride on the mechanism of protein fibrosis is beneficial to the development of protein fibrosis inhibitors,and has accumulated a certain theoretical basis for the development of neurological drugs.Through the exploration at the microbial level,it was found that BH has a certain inhibitory effect on the activity of multi-drug resistant bacteria.The research results of this paper re-understand the functions and effects of traditional drugs such as BH,and provide new theoretical basis and support for the optimization,development and clinical application of related drugs. |
