Bererbine hydrochloride(BER),as an inflammatory medicine,is widely used in clinical medicine.As one of the natural isoquinoline alkaloids,it can be extracted from the Berberidaceae and other Chinese plants.Bererbine hydrochloride was widely used for treating many kinds of diseases in clinical.It can even play a role in antimicrobial,hypolipidemic,immunomodulatory and so on.And in recent years,as a kind of feed and therapeutic drugs,berberine is widely used in animal husbandry and aquaculture.Too much berberine hydrochloride can't be completely absorbed.Most drugs and secondary metabolites are excluded in vitro.They contaminate groundwater and do great harm to human and environment.At the same time,large doses of berberine hydrochloride will kill microorganisms,destroy the ecological balance.Constitute a long-term threat to the ecological environment.Thus,in order to study the genotoxic and potential environmental of large doses of berberine hydrochloride.To evaluate the eco-toxicity of berberine hydrochloride in mouse liver cells,kidney cells,bone cells,spleen cells,lung cells and heart cells,biochemical responses including DNA damage and changes in the activities of antioxidative enzymes induced by berberine hydrochloride were investigated in mouse cells.In comet assay,use tail DNA content(%),tail length and tail moment,research the DNA damage in mouse liver cells,kidney cells,bone cells,spleen cells,lung cells and heart cells.By analyzing the activities of dismutase(SOD),catalase(CAT),peroxidase(POD)and malondialdehyde(MDA)to study the oxidative damage of berberine hydrochloride.Determine the residues of various organizations within the organs of berberine hydrochloride by UV spectrophotometry.The study is to probe the genotoxicit of berberine hydrochloride and to provide the theoretical supportion of follow-up study.In comet assay,we found that berberine hydrochloride can induce DNA damage on mice liver cells,kidney cells,bone cells,spleen cells,lung cells and heart cells.DNA damage in marrow cells had a significantly increase(p<0.01)after the exposure of different concentration of berberine hydrochloride.Berberine hydrochloride caused signifieantly increase of DNA damage in all treatments compared to the controls in a concentration dependent manner as observed in%DNA in tail,tail length and tail moment.SOD,CAT,POD and MDA are important enzymes in the anti-oxidant system of organism.In this study,the SOD,CAT and POD activities in marrow liver cells,kidney cells,bone cells,spleen cells,lung cells and heart cells were significantly stimulated in response to berberine hydrochloride in low dose(7.5 mg·kg-1)and then decreased to the levels of the control.It indicates that the cell has the capacity to tolerate the oxidative stress and activate the antioxidant system at the first stage of stress at low concentrations of berberine hydrochloride.However,with the concentration of berberine increasing(30mg·kg-1),the defensive ability of antioxidative enzymes would be lost.The enzyme activity was gradually decreased.Gradually increase the amount of drug residues in the range of 7.5mg·kg-1?60mg·kg-1.However,when the concentration of berberine hydrochloride increase to 120mg·kg-1,the value keep balance.The DNA damage and changes in the activities of antioxidative enzymes are related to the concentration of berberine hydrochloride in mouce cells.
