This experiment mainly pumpkin as raw materials,Bifidobacterium bacteria of pumpkin juice and preparation process of Bifidobacterium fermented pumpkin juice was optimized,and the annual processing factory of 3600 t fermentation pumpkin juice factory design,provides a theoretical basis for the development of the research results of pumpkin juice containing active probiotics.The main results are as follows:(1)With the clarification process of optimization of Pectinase on pectinase dosage,pH,temperature and time of the four aspects of single factor test and orthogonal test results show that the pectinase dosage is 1.6~2.2 g/L,pH 3.0~4.0,a temperature of 35~45 under the condition of pumpkin liquid clarification effect is good.The optimum conditions of orthogonal test,pectinase the amount of 2.0g/L,pH3.5,temperature of 35 degrees Celsius.(2)Through orthogonal test of sugar,honey and adding amount of sodium carboxymethyl cellulose(CMC-Na)effect on the taste of the product.The results showed that sucrose 6%,honey dosage 3%,CMC-Na dosage of 0.02%,the product is suitable,bright color,delicate taste,good stability,high sensory score.(3)The optimization of the fermentation conditions of pumpkin liquid of Bifidobacterium,found that with the prolonging of fermentation time,the number of bifidobacteria increased gradually,acidity increases with the prolonging of fermentation time,after 12 h,the acidity increased slowly and tends to be stable.The optimum process of Bifidobacterium fermented pumpkin juice was 3% inoculation amount,fermentation temperature 37 degrees,the fermentation time is 14 h,the product of bright color,good taste.(4)In this paper,the design of the factory with an annual output of pumpkin juice.The fermentation processing plant 3600 t from site selection,layout of the factory.The design of products and technology demonstration programs,quality standard,equipment selection,engineering,environmental protection,material balance,cost,profit estimates are analyzed.Prediction of production for a total investment of 47 million 217 thousand and 600 yuan,net profit of 15 million 839 thousand and 600 the yield reached 698 t yuan,can protect the business,operating rate of 73.5%,the payback period is 3 years.
