| Air-cured tobacco is variety,many of them have local characteristics,the development of Chinese-style cigarettes,especially the Chinese blended cigarettes,it is inseparable from the local air-cured materials.However,China’s air-cured raw materials exist poor quality,influenced by weather congenital problem,it can not be directly used as raw materials blended cigarettes,tobacco drying needs modulated [1].It is generally accepted that the enzyme and microbial played an extremely important role in fermentation process for tobacco.This study using traditional culture methods to study the Jiangxi "Black Tigers" air-cured surface of the microbial community structure combined with high-throughput sequencing methods,and isolated some microorganisms with enzyme production capacity.In addition,adding these microorganisms with enzyme producing ability to some low-grade tobacco,it’s supposed to improve the smoking of tobacco,and tap the potential of these microorganisms for industrial application.Research results consists of three parts:Combining traditional culture methods and high-throughput sequencing methods,microorganisms from surface of the four air-cured and sun-cured tobacco of Jiangxi "black tiger" samples via the two cultures(tobacco leaf liquid medium and solid plate culture broth,referred lc,pc),culturable microbes were sequenced and analyzed,and their community structure.The results show that after high-throughput sequencing analysis,microbial in four samples of tobacco leaf liquid culture are abundant than or flat broth culture."Black Tigers" air-cured surface there are three main categories,namely,Firmicutes(Firmicutes),Proteobacteria(Proteobacteria),Actinobacteria(Actinobacteria),the advantages of the phylum is Firmicutes and Proteobacteria.Isolated and identification microorganisms from the "Black Tigers" air-cured leaf surfaces of some enzyme,mainly screened for production of xylanase,cellulase,pectinase,amylase,protease,to give a total of 85 strains of bacteria.It was found that these bacteria produce xylanase,pectinase,protease relatively strong capacity,followed by amylase,cellulase finally.Some bacterial of strong enzyme production was identified as Bacillus pumilus,Bacillus subtilis,Bacillus methylotrophicus.The preparation of enzyme production of bacteria and flora into microbial agents,its viable count reached 109-1011 cfu / g,still with corresponding enzyme production capacity.These agents added back to the low-grade Jinsheng cigarette tobacco group leaves and unblended tobacco leaves for aging,smoking by experts for sensory evaluation and gas chromatography-mass spectrometry to analyze the aroma-producing substances and irritating harmful substances changes after added agents to aging.The results found that some agents can significantly improve the properties of tobacco smoking,sensory evaluation score increased by more than 13.5 points,GC-MS showed that the production of incense material production,while reducing the content of harmful substances.These results provides an important basis of digging air-cured surface of microbial resources and their application. |
