After harvesting, baking and repeatable flue-cured, the quality of flue-cured tobacco is still far from enough good yet. Aging process is still needed to make tobacco soften to meet the requirement of processing. Nature aging is mainly the method, but it needs 2-3 years, impropriates much storage and costs a lot.So, it has great economy value to study on the varieties of enzyme and microbe during tobacco aging and then develop bio-agent for shorting aging period, improving tobacco quality.(1) The results of microbe's isotation and primary identification during aging stage indicate that microbe is active and the microbial species include bacteria, actinomycetes, and moulds.(2) During the nature aging of flue-cured tobacco, which proteinase and a-amylase show the great change in activity, indicates that they take part in the fermentation and really make a difference especially.(3) In present study, through primary selection and orthogonal cross-course method, we confirm the optimum combination including accelerant 0.09%(dry weight of the leaf), 2# pectic enzyme 0.2%, 6,γ-xylanase 0.2%, Neutrase (Novo Nordisk) 0.2%, mixed ion 0.03% from twenty kinds of enzyme and chemistry agents.(4) Using our microbial agent in smashed and repeatable flue-cured tobacco C3F of Sichuan province, the results shows the content of total nitrogen, total sugar, protein, amino acid and nicotine decrease in the aging stage. Compare to nature aging, the decrease extend is bigger, especially in the first 1-6 months. This phenomenon indicates tobacco fermentation is intense with microbial agent. After that, change is slow down and majority aromatic components increase with different extent.(5) We get seven strains producing fragrant at last and use them in fermentation of tobacco extraction to prepare tobacco-improving agent added other natural plant extraction. Dealing with tobacco with this agent, tobacco smoke is soft, aroma quality is better and peculiar smell, thrilling is light.
