Herba Pteridis multifidae is affiliated to the all dry grass of the Pteris multifida Poir. in Pteridaceae,another name pheasant-tail, the grass on the brandreth or iron-chicken,etc. Pteris multifida Poir. is a Chinese herbal medicine that it has various effects and abundant resources, a long history to be used.Therefore, it is great significant to study and exploit Pteris multifida Poir..In this study,the all goal is to study on extraction and purification technology of the total flavonoids and the effect of treating scald in Pteris multifida Poir..We also have another goal to lay the foundation for the exploition and utilization on Pteris multifida Poir..All of the results show as follows, respectively.1. To optimize the extraction of total flavonoids in Pteris multifida by central composite design-response surface methodology. METHODS:With Ultrasonic wave extraction, the independent variables were ethanol concentration(%),extraction time(min) and liquid-to-solid ratio(times), the dependent variable was the extraction rate of total flavonoids in Pteris multifida on the basis of the single factor experiment of influencing factors.Linear or second-order quadratic models were used to estimate the relationship.Central composite design-response surface methodology was used to optimize the process of extraction. RESULTS:The optimum extraction conditions were as follows, 63.7% ethanol concentration, extraction time for 25.8 minutes, and liquid-to-solid ratio was 44.7 times.Under these conditions, the experimental rate was 1.814%, bias between predicted and observed of rates was -0.928%, close to the predicted value 1.831%. CONCLUSION:The experimental design is simple, reliable and highly predictive.2. To primarily study the purification technology of total flavonoids from Pteris multifida. Taking total flavonoid content of Pteris multifida as the index, to determine the best resin for the adsorption and separation of the total flavonoids of Pteris multifida,through six kinds of different types of macroporous resin filter and to determine the resin separation and purification total flavonoids conditions of Pteris multifida by the single factor. Result:The adsorption and separation performance of AB-8 resin is very perfect in the total flavonoids of Pteris multifida, the adsorption and separation process conditions of the total flavonoids:the best technology of the total flavonoids of Pteris multifida is that the extracted liquid (2.07mg/ml) 10ml through AB-8 resin column, adsorption 30min, elution with water to remove impurities, using 90% ethanol 120ml (elution the speed 2ml/min) elution, the resin can be reused four times. ConclusionIn:Effect of purified by AB-8 resin Pteris multifida total flavonoids is obvious.3. To observe the therapeutic action of extract from Pteris multifida Poir. on the laboratorial scald model of wisterRats. WisterRats were scalded to thermostatic waterbath at 85 ? for 8s to produce second-shallow wounds and were divided into five groups.Three groups were respectively treated with the extract from Pteris multifida Poir. of high?middle and low dosage and the positive control group with Jing wanhong ointment for 25 days.The changes of scald wounds area and tissue pathological slice were compared every 5 days.We also study the tissure section of scalded tissue.Result:The scald wounds area in high and middle groups were significantly less than that in the positive control group since the 10th day (p<0.05) , the healing effects in all dosage groups were obviously better than that in the positive control group. Conclusion:The extract from Pteris multifida Poir. showed remarkable therapeutic action on the laboratorial scald model of wisterRats.
