| In this paper, extraction and purification process of total flavonoids from hawthorn leaves were studied. Getting a rational technological process. Establishing the foundation for the industrialization.The experiments focus on three kinds of extracting methods : water extraction, enzyme extraction and ethanol extraction. The optimum extraction condition with water is described as following : extraction temperature is 90℃, extraction time is 90min , pH of extraction media is 12, and the ratio of solid to solution is 1:20. The optimum ethanol consistency is 70%. Orthogonal test table L18(37) was used on the base of single factor test. The optimum technology for enzyme extraction was showed as following : The temperature of enzyme degradation is 60℃, the time of enzyme degradation is 20min, pH of extracting media is 5.0, enzyme consistency is 0.15mg/ml, the time of extracting with water is 90min. Among the three kinds of methods, the result shows that: extracting total flavonoids from hawthorn leaves with cellulase is the most rational. Compared with traditional extraction technology with water , the extracting ratio of total flavonoids is increased by 18.59%.The comparision has been made between the two methods of removing the impurity with clarifying agent and ethanol agengt. Some affecting factors such as the joining order of clarifying agent;the amount of clarifying agent , reacting temperature , reacting time , the consistency of solution etc have been studied. The result indicates that clarifying agent is superior to ethanol agent. The optimum conditions for the purifing technology with clarifying agent are as follows : B component is added first and then the A component is added subsequently, the amount of B is 1.2g/L, the amount of A is 0.6g/L,the reacting temperature is 40 ℃ , the reacting time is 40min, and the exacting solution is diluted adequately.The total flavonoids of hawthorn leaves were purified with macroporous resins. The optimum macroporous resins absorbing-sucking conditions obtained were: the solution temperature is 40℃, the consistency of solution 0.75-1.0g/L, pH of solution media is 4.5, the consistency of eluting solution is 80% ethanol solution , the amount of elution is 3BV, and the speed of eluting is 1.5BV/h. |
