| This paper aims to develop an improved studies of extraction, purification and antioxygenic ability of Calycopteris floribunda (Cf.). It provides a theoretical basis for processing of Cf. flavonoids, maximizing its utilization and economic benefits.The main results obtained from the experiment were as follows:(1) The UV-Vis was used to determine total flavonoids in Cf. at the detection wavelength of410nm; The HPLC was used to determine the pachypodol, wavelength is365nm, temp is35℃, flow rate is1mL/min, mobile phase is CH3CN-H2O (75:25). Through the methodology test, this method is simple, sensitive and accurate in operation and good in repeat. It is suitable for the determination of total flavonoids and pachypodol in Cf..(2) The single factor experiments and center composite design have been used to optimize the extraction processes of total flavonoids of Cf. The optimum extraction conditions are as follows:extraction time is15h, temperature is76℃and ethanol volume fraction is55%. Under these conditions, the yield of Cf. total flavonoids is2.61%, consistent with the prediction value2.63%.(3) To investigate the optimal purification process of the total flavonoids from Cf..4types of macroporous resin are used to study the static and dynamic adsorption performance of total flavonoids, of which key factors were investigated, including sample pH, sample mass concentration and volume fraction of desorption solution. RESULTS: The HPD-450macroporous resin shows best adsorption and separation performance. Freundlich model is better than Langmuir model for describing the adsorption of flavonoids. The optimal adsorption condition is as follows:sample pH is5, sample mass concentration is0.6-0.8mg·mL-1, sample volume is3BV (resin bed volume), the eluent volume is3BV, consisting of70%(mL·mL-1) ethanol. CONCLUSION:The mass fraction of flavoniod is75.86%which shows a significant increase than20.23%before purification, and the recovery rate of total flavonoids was62.65%. (4) The pachypodol of separation process by AB-8resin were as follows:extraction solvent is ethyl acetate; sodium carbonate solution of pH gradient extraction; elute solvents are water and25%alcohol to remove polar components;70%alcohol is used to obtain the pachypodol after pH value of the elute solvents was adjusted to5by5%hydrochloric acide; the pachypodol was crystallized under low temperature under Ethyl acetate-petroleum ether system. Under these conditions, the purity of pachypodol was96.4%.(5) Five compounds’ ability to remove superoxide radical system and hydroxyl radical system were tested, these five compounds are crude Cf. flavonoids extraction, refined Cf. flavonoids extraction, Vc, rutin and pachypodol. |
