To study on extraction,purification and stability of lycopene,fresh tomatos were used as raw material. The purpose was to improve the extraction yield of the lycopene,and improved the purity of product and the stability of lycopene. Lycopene purification part used column chromatography and crystallization to determine the conditions of column chromatography, the ways and conditions of crystallization.The part of stability developed the lycopene microcapsules and microemulsion, and chose the optimal process conditions, and the stability of finished product testing, mainly the following results:Using ultrasonic assisted extraction, extraction conditions were material liquid ratio of1:5, under the condition of ice bath extraction for 15 minutes, and ultrasonic power was390 w. After the extraction, the lycopene concentration was 92.66 ug/ml.The experiment of column chromatography purification condition determined D3520 macroporous adsorption resin column chromatography as adsorbent and 95% ethanol as the solvent adsorption. It determined the best adsorption material concentration of 0.00023 g/m L, and adsorption temperature of 30 ?, and the adsorption rate and elution rate of 3 m L/min. The elution mode was step by step, after the first ethyl acetate ethyl acetate: chloroform was 2:1, the eluent containing lycopene content was 80.46%, yield of 25%, the purity of the product increased by 3.91 times.Crystallization purification experiment used orthogonal experimental design, and optimized the parameters of saponification tomato red oil dementholized resin to get the best material ratio of 1:1.2, the reaction temperature of60 ?, the reaction time of 80 min. Choose ethyl acetate solvent crystallization,crystallization temperature of 0?, the crystallization time of 2 hours, and crystallization series of secondary, to get acicular, deep red crystal. The product was checked by high performance liquid chromatography(HPLC).At last, the purity was 82%,and yield reaches30%.The best technology to optimize the conditions of lycopene microcapsules followed the next conditions. Core material joined in the form of solution, and it used gelatin + sugar+?-cyclodextrin as wall material. The core material of total material radio was 1.2 mg/g,aperture size with 10 ml syringe, latex temperature 30 ?, emulsion viscosity to add 10 ml of distilled water, curing agent to starch. Preparation was good lycopene microcapsules with aromatic flavor, smooth the surface of spherical particles in orange color. Particle diameter was 2.26 mm, water content was 33.5%, the density of 1.1 g/ml, solubility of88.87%, the surface of the microcapsules lycopene content was 5.8%, the yield of38.8%.Preparation of lycopene microemulsion, through experiment data quasi ternary phase diagram, finally confirmed microemulsion formula: oil phase for lycopene crudeextract, ethyl acetate, anhydrous ethanol.The proportion of Lycopene crude extract and ethyl acetate was 1:12.5.The proportion of the sum of lycopene crude extract and ethyl acetate and anhydrous ethanol was 1:1.The emulsifier was twain- 80, and the proportion of oil phase and twain- 80 was 1:0.4. The water phase was anhydrous ethanol and distilled water. The proportion of anhydrous ethanol and distilled water was 1: 1.The experiment finally selected for crystallization purification method, method to improve the stability of microemulsion method choice.These results provided a theoretical foundation for industrialized production of lycopene product.
