The Role Of The Sperm-specific Lactate Dehydrogenase In Ochotona Curzoniae Tissues

Plateau pika?Ochotona curzoniae?has very strong hypoxia adaptation.Previous studies found that the sperm-specific lactate dehydrogenase?LDH-C₄?gene is expressed in Plateau pika somatic cell.Intraperitoneal injection is a simple and convenient method to drug injection,in order to shed light on the role of LDH-C₄ in plateau pika tissues and verify the silence effect of Ldh-c in Plateau pikas by intraperitoneal injection of adenovirus pMultiRNAi-Ldhc with the mothod of RNA interference,we silenced the Ldh-c gene in Plateau pika heart,live,brain and skeletal muscle by RNAi;27 Pikas were randomly divided into the RNAi-LDHC,RNAi-HK and Control,each group has nine individuals.The Pikas of RNAi-LDHC,RNAi-HK were injected with 0.65 mL adenovirus pMultiRNAi-Ldhc and pMultiRNAi-NS,respectively.The Control group were no injected.Seven days after injection,the expression level of Ldh-c mRNA and protein in Plateau pikas heart,live,brain and skeletal muscle,and the determination of the lactate dehydrogenase?LDH?,lactic acid?LD?and the content of Adenosine triphosphate?ATP?were detected.The results indicated that intraperitoneal injection of adenovirus pMultiRNAi-Ldhc could significantly reduce the expression of Ldh-c in tissues of Plateau pikas.Specifically,in the mRNA and protein levels,the expression levels of Ldh-c in heart were decreased by 48.11%and 19.27%;in liver were decreased by 70.16%and 25.82%;in brain were decreased by 49.08%and 25.36%;in skeletal muscle were decreased by41.73%and 15.76%,respectively.Meanwhile,when Ldh-c gene expression was silenced,the activities of LDH,the contents of lactic acid and ATP in each tested tissues were generally decreased by 25.58%,41.94%and 21.23%;28.16%,15.90%and 24.66%;16.65%,12.78%and 18.50%;23.98%,51.08%and 19.29%,respectively.It indicated that intraperitoneal injection of adenovirus pMultiRNAi-Ldhc can effectively silence Ldh-c in Plateau pika.Under the condition of severe hypoxia,the heart,liver,brain and skeletal muscle tissues of Plateau pika can generate part of ATP upon LDH-C₄ catalyzed anaerobic glycolysis and therefore enhance the host adaptation to the hypoxic environments.
