Sika Deer(Cervus nippon)and Red Deer(Cervus.elophus)are a kind of protected animals of the national grade I and grade II,not only has high economic value, but also has important ecological protection value.Due to the increasingly serious destruction of habitat, the number of Wild Sika deer and wild Red deer decreased rapidly. In recent years,it has been found that there is overlap between the Wild Sika deer and wild red deer in Northeast china. How to study and protect the two populations has become the focus of future research. This study was conducted to study the genetic diversity of wild wapiti and sika deer in Northeast china. We collected 240 samples of suspected Wild Sika deer and wild red deer in the Laoyeling area of the Wangqing Forestry Bureau in Jilin province and the Muling Forestry Bureau in Heilongjiang province from January to December,2015 and 2016.The DNA was extracted by Qlamp DNAstool Mini Kit in stool DNA kit.In this paper, using the cyt b gene sequence analysis of mitochondrial DNA in the Northeast Chinese acquisition of suspected wild deer and suspected wild wapiti fecal samples belong to sika deer and red deer, microsatellite markers for individual identification of wild deer and wild red deer samples, differences in genetic diversity using mitochondrial DNA cyt b gene, microsatellite and wild deer wild red deer,The results are as follows:1.There are 240 samples are determined through comparison with the sequences of Cty b gene from blast in the Gen Bank database. There are 23 samples of Sika deer and 37 samlpes of Red deer.2.The 9 pairs and 12 pairs of microsatellite primers were used to identify sika deer and red deer.23 samples of Sika Deer feces were obtained from 17 individuals, and the samples from the 37 red deer were obtained from the 22 individuals.3.There were 6 haplotypes in Wild Sika deer, the population haplotypes of wild red deer were 5.Haplotype diversity and nucleotide diversity were lower in haplotype diversity and nucleotide diversity of the Northeast subspecies than those obtained by other researchers.
