Panjin Red Beach is a unique wetland resources in Liaoning province.It keeps its unique natural appearance because it was rarely influenced by artificial condition.There is in plenty of microbial resources in Panjin Red Beach,it will make contribution to discover new microbial resources and internal soil microbial community composition through investigation and research on the soil microbes,using biological methods to screen microbial resource and the microbial diversity.It is also of great significance to explore the origin of life and evolution,as well as the mechanism of microorganism and environmental interaction.In this study,parts of halophilic and alkalophilic bacteria of Panjin Red Beach were screened and microbial diversity was studied using highthroughput sequencing analysis.One of high yielding alkaline protease producing strains was selected to preliminary study the enzymology characteristics.The main results were as follows:1.5 groups of 15 soil samples from Panjin Red Beach were analyzed by highthroughput sequencing technology.The results showed that the bacterial community of Panjin Red Beach was mainly composed of 9 phylum,among which proteobacteria(45.24%),Bacteroidets(21.90%),Firmicutes(8.75%),Chloroflexi(6.92%),Actinobacteria(4.32%),Gemmationadetes(3.09%),Acidobacteria(2.48%),Planctomycetes(1.63%),Ignavibacteria(1.05%),others 4.62%.2.Analysis on cultured microbial community composition in saline-alkali land of Panjin Red Beach showed that 80 halophilic and alkalophilic bacteria were isolated by pure culture.It can be seen from the strain classification information that 80 strains were belonging to 10 genera and 7 phylum,among which Bacillus was the dominant populations.3.A high yielding alkaline protease strain HHT597 was isolated and screened from soil,the molecular biology identified result showed it was Bacillus altitudinis and halophilic and alkalophilic bacteria as the optimum pH value of strain HHT597 was 8.5,it grew well in 2 % of NaCl.The resistance of strain HHT597 was proved that it could grow in extreme circumstances.It could also produce alkaline protease under alkaline conditions.It was possible that strains HHTT597 could provide related enzymes under alkaline conditions for industrial production.4.The alkaline protease got from the experiment had good resistance to salt,a good stability of 60 ? high temperature and still did well in enzyme activity under the condition of high salt concentration,owned good characteristics on decomposable the of the protein at three kinds of substrates.Therefore,it shows great application value in industry.
