Nostoc commune Vanch popular named Nostoc commune, lichen etc. it is belong to Cyanophyta. Cyanophyceae, it is the frond of belonging to Nostocaceae, nostoc. Nostoc commune Vanch been called nostoc commune usually, because the boundedness of wild Nostoc commune Vanch, it is needs rigorous condition and sensation to the pollutant. So the wild Nostoc commune Vanch is tapering off. The aritificial liquid culture of Nostoc commune Vanch not only protecting and utilizing itself but also having a well foundation to the deep processing of Nostoc commune Vanch. At the same time, it has a remarkable significance to the economic growth of diversificative economic and exploring local financial condition. The PC containing in the Nostoc commune Vanch not only having various physiological activity but also having the nice expression in antitumor, anti-inflammatory, antioxidant. And the fluorescent characteristic of Nostoc commune Vanch also having the important and practical value in making fluorescence probe.This article takes wild Nostoc commune Vanch as the raw material, devising experiment and searching the condition of culturing aritificial liquid Nostoc commune Vanch. At the same time, this article researched the technology of extracting Phycocyanin and the technology of detached Phycocyanin. This article obtains the following achievements:1. According to the literature and experiment, setting up the sterile culture system of Nostoc commune Vanch, screening out the most suitable liquid nutrient medium for the Nostoc commune Vanch is Zhu’s10culture solution. Proceeding the four factors and three levels orthogonal test basing on the single factor testing of artificial cultivate Nostoc commune Vanch. According to the results, the optimal formulas is:Na2CO30.025g/L, MgSO4·7H2O0.02g/L. Na2SiO3·9H2O0.04g/L, K2HPO4·3H2O0.02g/L, Ca(NO3)2·4H2O0.05g/L, FeCl3·6H2O0.025g/L, VB0.5g/L.2.The experiment confirms the optimal no-nutrition condition in the liquid culture of Nostoc commune Vanch is:the optimum is pH8, the optimum temperature is25℃S.Acconding to comprehensive research, the way to purification of detached Phycocvanin is:soaking the Nostoc commune Vanch in phosphate buffer12hoursâ†'multigelation7timesâ†'ultraphonic extracting5timesâ†'centrifuging and collect supernateâ†'air pump filtrationâ†'30%salting out the useless proteinâ†'60%salting out and precipitating phycocyaninâ†'dialyzing desaltingâ†'DEAE-cellulose ion exchange column, and collecting the purer phycocyaninâ†'sephadex G-150gel columnâ†'getting the purer phycocyanin. The purity of final phycocyanin getting from experiment is4.55. accord with the standard (purity>4).4. This experiment contrasts the content of phycocyanin between wild Nostoc commune Vanch and the aritificial liquid culture of Nostoc commune Vanch, at last, we find that the phycocyanin in wild Nostoc commune Vanch is higher than aritificial Nostoc commune Vanch, according to the experiment, maybe the difference been aroused by different form. We hope to improve the content of phycocyanin in aritificial Nostoc commune Vanch from increasing the time of cultivating Nostoc commune Vanch.
