Plant growth process is affected by a variety of abiotic stress all the time.Most of the economic crop yield reductions resulted from abiotic stresses,so the research of abiotic stress on plant has very significant economic benefits and academic value.In recent years,many abiotic stress relatedgenes have been identified in Arabidopsis thaliana,but plant resistance to stress mechanism is very complex,there are still many problems to be solved.In order to identify new abiotic stress related genes,we integrated the large number of existing bioinformatics information,and screened a number of abiotic stress related genes.After several screening work we determines the candidated genes.We use the T-DNA insertion mutants as materials,and study mutant according to the prediction information on network.In order to get obvious phenotype mutants,then get abiotic stress related genes,and study their role in abiotic stress response.Screening out 27 genes,including low temperature,ABA,osmotic,salt and drought stress related genes.T-DNA insertion mutants were signed from Arabidopsis seeds resource center.Then screening 22 homozygous lines were identificated.One of the most important problems on the currently study of low temperature stress is the reception of the low temperature.After cold shock,[Ca2+]cyt rise in plant cells,on time and mechanism is closely related with the process of low temperature stress.So the research for the low temperature stress related mutants is mainly on detect the change of [Ca2+]cyt.The mutant was transformed by aequorin,and detected the change of [Ca2+]cyt under cold shock.The changes of [Ca2+]cyt were similar in mutants lines,no significant differences.The Ca2+-mediated signaling system in the low temperature transduction pathways is in a upstream position,the data we have presented show that the gene screened out may not be directly involved in the induction cold feeling Ca2+ signal,or do not directly control the Ca2+ signal induced by cold.For other abiotic,osmotic,salt,drought and ABA related mutants,the root elongation experiment results show that atsrg5 more sensitive to NaCl than wild type,and the lines atarg10 and atarg19 are more sensitive to ABA than wild-type.The further study of atarg10 shows that the expression of AtARG10 is not only regulated by ABA but also regulated by low temperature,salt stress.The germination experiments shows that the atarg10's germination rate is higher than the wild type in NaCl treatment.The cotyledons green rate is obviously earlier than the wild type show atarg10 affaiblir the inhibition effect from NaCl on Arabidopsis thaliana germination and growth of seedlings.AtARG10 expressed in flower is high.atarg10 bolting is earlier than the wild type,and the number of rosette leaves are smaller than the wild type.At present,AtARG10::super1300+-GFP expression vector has been successfully constructed.The super expression vector,GUS vector,and the instantaneous short carrier are building.
