Six kinds of leopard cat tissue were selected to undergo primary culture.Three kinds of cells:chondrocyte-derived cells,heart-derived cells and lung-derived cells were successful.These cells were used to research the biological characteristics of leopard cat cells.In this study,the following experiments were carried out:primary cell culture,morphology observtion of subcultures,statistics of cell adherence rate,comparison of the rate of cryopreservation,comparison of three kinds of cell growth curves,detection of mycoplasma contamination,karyotype analysis and so on.In morphological observation,the three source cells all belong to fibroblast-like cells,but each of them has significant characteristics.Adherence rate analysis datas showed that the heart-derived cells were lowest and chondrocyte-derived cells were highest.The lung-derived cells were easy to adhere before P9 but these cells were difficult to adhere and grow after P9.The survival rate of these three kinds of cells showed that:after passaged several tims,survival number declined in all cells.The cell curve showed that the curves were shown as "S",and the chondrocyte-derived had a fastest proliferation,followed by the lung-derived cells and heart-derived cells.In addtion,we did not find any mycoplasma in all cultured cells.In leopard cat cell karyotype analysis,we found that the number of chromosomes was 2n = 38,and 18 pairs were autosomes.The morphological type was 7M + 11SM,and 1 was sex chromosomes.The morphological type X was ST.This study provides a good basis and experimental material for research biological characteristics and preservation of genetic information of leopard cat cells in future.
