The effects of Human-like Collagen-I (HLC-I) on cell growth were studied by BHK-21 cell in vitro. The following works have been done during the study.1 The effect of HLC-I on BHK-21 cell growthThe growth curve of BHK-21 cell was created by cell cultured in vitro. The effects of various Human-like Collagen-I concentrations (0.1‰, 0.3‰, 0.5‰, 0.7‰, 0.9‰, 1.1‰, 1.3‰) at different cultivation time (24hr, 48hr, 72hr, 96hr, 120hr) on cell growth, proliferation and viability were studied by MTT Assay, Try pan Blue Dye, cell counting and statistic analysis. The present study showed that RPMI1640 media in the presence of Human-like Collagen-I concentrations at 0.3‰, 0.5‰, and 1.3‰ had significant influences on cell growth (P<0.05) , 0.3‰ and 0.5‰ of Human-like Collagen-I in media had positive effects on cell growth and proliferation rate, while 1.3‰ of human-like collagen-I in media had negative effects. Various Human-like Collagen-I concentrations had no significant influences on viability rate by Try pan Blue Dye (P>0.05) . Human-like Collagen-I also had positive effects on cell division rate after cultured 48hr. It demonstrated that Human-like Collagen-I had positive effects on BHK-21 cell growth at lower concentrations and no significant influence on viability rate thus showed good biocompatibility.2 The mechanism of HLC-I on BHK-21 cell growthThe effects of HLC-I on cell proliferation, cell apoptosis rate, G1%, G2% , S% of cell growth and morphology at 24 hr were studied by MTT Assay, LDH analysis, FCM and morphology. The effects on BHK-21 at 24 hr showed that RPMI1640 media in the presence of Human-like Collagen-I concentrations at 0.2‰, 0.3‰, had positive effects on cell growth (P<0.05), but had no significant influences on LDH content in culture media. FCM analysis demonstrated that HLC-I (0.1‰, 0.2‰, 0.3‰, w/v) made a slight change on cell apoptosis rate, and HLC-I had no significant difference between cell growth in G1%, G2% , S% and that of blank control. Morphology observation showed that HLC-I (0.3‰) elongated cellsand made them become slender.
