Since transistor circuit approaching its performance limit, working frequency and calculation speed of traditional electronic computer are also limitted. Recent years, scientists have begun to explore a new calculation model instead of traditional electronic computing. The DNA computer has been heavily favored by the scientific community due to its advantages such as high parallelism, mass of storage capacity. Logic operation and arithmetic operation are the most basic functions of computer operation system, therefore, the research of logic and arithmetic operation in DNA computing is the key to realization of DNA computer functions. DNA computation can not separate itself from traditional biochemical reaction, however, the traditional biochemical reaction has its disadvantages such as of bulky of equipment, complex process, and difficulty of control. Using microfabrication technology to prepare various modules on microfluidic chip for implementing biochemical reaction not only has its advantages such as small volume, convenience of carry and less sample consumption, but also provides powerful technical support for the design and implementation of large-scale practical DNA computers. Therefore, the research of DNA computing system based on microfluidic chip has become the new hot spot.In this paper, molecular beacon replaces DNA molecule as the carrier arithmetic operands, and the idea of DNA computing system is combined with microfluidic technology to design microfluidic chips which use molecular beacons to complete logical operations and N-bits binary arithmetic operations. And it also illustrates method and process of operation in the chip to complete the calculation in details. On this basis, the temperature control system which is need for the chip to complete the calculations is designed, and this paper briefly introduces the overall structure and software programming environment of the system, and the specific design method and function implementation results of its power supply module, temperature acquisition module, temperature control hardware module are presented. In order to real-time control the temperature, RBF neural network PID control algorithm is adopted to track temperature error signal. And the traditional digital PID algorithm and RBF neural network PID algorithm of tracking unit step signal are simulated in MATLAB environment, the simulation results show that the RBF neural network PID control algorithm has a better tracking response effect. Finally, a fluorescence detection and processing system for detecting the calculation results is designed and the overall structure of the system is given. According to the mechanism of fluorescence, right components are chosen to design fluorescence detection system and basic circuits of signal processing system, and a PC control interface of the detection processing system is realized by programming using Visual Basic software.For a long time, most scholars devoted themselves to model research of DNA computing, while the research of its design and implementation is less, and the implementation of DNA computing through biochemical reactions has been faced with the problem that final solution is difficult to detect. Though information processing by controlling flow of controllable fluids in microfluidic chip is still in its infancy, it is likely to become an important branch in the field of information processing in the future, and its potential and influence should not be ignored. This paper solves the problem of difficulty in operation result detecting by adopting molecular beacon instead of DNA molecules as arithmetic operands, and it realizes DNA computing through biochemical reactions on microfluidic chip, which not only overcomes the weakness in traditional biochemical reactions, but also pushs DNA computing from theory to realization. Therefore, research of DNA computation based on microfluidic chip has important theoretical significance and practical value for realization of DNA computing theory and exploration and development of DNA computer.
