Objective: This paper main to study and research the expressions of interferon-γ(IFN-γ) and interleukin-4(IL-4) specific receptor mRNA in spleen of mice in period offatigue. To analysis the dynamic changing features of the IFN-γ/IL-4mRNA andrelated parameters in spleen of mice during different phases of recovery period offatigue. Explore mechanism of mobility immune suppression in period of fatigue,acquire to defer the development period of fatigue and maintenance the method of thedynamic changing after period of fatigue. To effectually and feasibly provide theoryaccord which immune of dynamic changing.Methods:48female SD rats (2month old) were randomly divided into silent group (Sgroup, n=16), exercise groups (E group, n=16), cordyceps militaris+exercisegroups(C+E group, n=16). At the sampling time, the S, E and C+E groups wererandomly divided into the group after exercise36h S1group(n=8), E1group(n=8),(C+E)1group(n=8), and one week S2group(n=8), E2group(n=8),(C+E)2group(n=8)after exhaustive running. S group unaffectedly growth in cage, all rats in experimentalgroups were assigned a9-week training program increasing load by degrees with a finalexhaustive running. And the E group was taken Sodium Chloride0.9%2ml byintragastric administration, C+E group was taken Cordyceps Militaris2ml (400mg/kg)by intragastric administration. The serum WBC, RBC, Hb, CK, BUN and T weremeasured from the blood samples after36h of first and third week exercise. S1group、E1group and (C+E)1group were peritoneal injection1%pentobarbital(1ml/100g weight)to test index same as above, test the expressions of IFN-γ and IL-4mRNA from spleenof mice. S2group, E2group and (C+E)2group were test index same as above after oneweek. The statistical results of all data was representation of average value±standarddeviation (x±SD). S1group, S2group, E1group, E2group,(C+E)1group,(C+E)2groupwere analysis by One-Way ANOVA, significant difference for p<0.05, everysignificant difference for p<0.01.Results: All rats in S1, E1and (C+E)1groups were assigned a9-week training programincreasing load by degrees, the change of biochemical indicator after36h.(1)The E1, (C+E)1of serum Hb and T were significantly lower than that of S1, respectively(p<0.05). The E1,(C+E)1of serum CK and BUN were significantly higher than that ofS1, respectively (p<0.05).(2)The E1,(C+E)1of serum WBC were significantlylower(p<0.01), and (C+E)1of serum WBC was significantly higher than that of E1,respectively (p<0.05).(3) The expressions of the E1which IL-4and IFN-γmRNA weresignificantly lower(p<0.01), and IL-4/IFN-γmRNA was significantly higher. Theexpressions of the (C+E)1which IL-4and IFN-γmRNA were significantlylower(p<0.01), and IL-4/IFN-γmRNA was not significantly difference. The proteinumexpressions of IL-4and IFN-γ were significantly lower (p<0.05). After one week ofexhaustive exercise, the IL-4and IFN-γmRNA were nearly that S2group, Theproteinum expressions of the IFN-γ that E2and (C+E)2groups were significantlyhigher(p<0.05).Conclusion:(1)The rat was instantly reach fatigue after treadmill movement through9weeks, compound Cordyceps militaris can retard significantly movement fatiguehappen and anti-fatigue.(2)The rat was instantly reach fatigue after treadmill movementthrough9weeks, the immunifaction WBC of rat was inhibited significantly, destrpy thedynamic balance of Th1/Th2, but the compound Cordyceps militaris can improve theinhibited effective of immunifaction, and that retain the dynamic balance of Th1/Th2.(3)After movement through9weeks, the Th1/Th2that body immunifaction retain thedynamic balance after one week reablement. Exercise-induced Immunosuppression wasprovisional and reversible process.
