Ephedrine,(1R,2S)-2-dimethylamino-benzene propane-1-alcohol, a kind of alkaloid, is exist in the plant ephedra. Ephedrine can be used to treatment the allergic diseases, such as: asthma, hay fever, whooping cough etc. Andit has excitatory effects on the central nervous system, is similar with amphetamines. Ephedrine is not only a kind of stimulant drug, but also a raw material of methamphetamine. Therefore, the regulations be published to tightly controlled the use of ephedrine, for instance: The IOC-MC banned athletes use the drug that contains ephedrine during the game; In 2005, China promulgated the “Regulations on the administration of precursor chemicals”, ephedrine and its salts are defined as controlled drugs; the revision of the "dangerous chemicals management regulations", the ephedrine is listed as controlled substances; In 2012, The State Food and Drug Administration confined that the minimum content allowed in ephedrine, and points out that compound preparations of drugs registration be approved. Furthermore, the real identity should be used to buy the drugs which contain ephedrine.At present,the researches mostly focus on detect ephedrine in drugs. And the determination methods of it in complex biological samples, is less reported. The analysis of ephedrine in complex biological samples is studied by GC-MS in this paper.To developing a new analysis method of ephedrine, some regents and conditions are examined. For example: internal standard, derivatization reagent, derivatization methods, derivatization reagent dosage, the derivatization time, chromatographic column, temperature program and mass spectrometry conditions. A GC-MS method is established for detecting the ephedrine in blood and liver of mice. Experimental results show that, the best conditions is: Elite-35 MS quartz capillary column(30m×0.25 mm, 0.25μm), the injection temperature is 270℃, the carrier gas is helium(99.999%), the flow rate is 1.0m L/min and constant current mode, programmed temperature: 80℃'12℃/min'220℃(5min)'12℃/min'280℃(5min), Sample volume: 1μl.EI source, 70 ev, 40-500 amu, ion source temperature: 250℃, transmission line temperature: 250℃, The results for the analysis of ephedrine in the blood is: linear regression equation:Y=1.67326X+35.6341, linear range: 50~500ng/m L, detection limit: 11ng/m L, relative recovery: 89.47~104.43%. The results for the analysis of ephedrine in the liver is: linear regression equation: Y=1.67326X+35.6341, linear range: 50~500ng/m L, detection limit: 17ng/m L, relative recovery: 97.89~102.13%.Using the GC/MS method established in this paper, the blood, liver and hair samples of mice are analysed.The content of ephedrine, standard deviation, and other results hair showed that the method can be used in the analysis of trace ephedrine in complex biological samples, and the effect is good.
