| Objective:To Study the extraction process and purification process of flavonoids of Sarcandra glabra.Method:With the measurement of flavonoidsã€Isofraxidin and extracting ratio as index, the optimum extraction process was selected with the orthogonal design method.With the measurement of flavonoidsã€Isofraxidin and extracting ratio as index, the first process of optimum purification condition comfirmed by uniformity design. During the reseach of purification with rosin,with the ratio of mobile-absorbing and separating as index,The optimum parameters of purification were determined by singal factor. Using the content of flavonoids as a index, the effect of different temperature was contrasted after vacuum drying.The quality standard of Sarcandra glabra was studied by TLCã€visible spectrophotometry and HPLC.Result:(1) extraction process Sarcandra glabra was crushed into rough powder,2 times of refluxing with water,14ã€12 times of water-adding amount and 2.5,2.0 hours of refluxing time respectinely. (2) purification process The alcohol method was Comfirmed to use to flocculate the water solvent and the optimum purification condition was:relative density of concentrate was 0.6(meterials vs extracting solution),the percent of alcohol was 50%,the temperature was 20℃. The optimum parameters of purification was:sample concentration was 0.6g·ml-1,sample flow velocity was 2BV·h-1,then being eluted with volume of 4 BV of water, volume of 4 BV of 40% Etoh at the velocity of 16BV·h-1 gathering the part eluted by 40% Etoh. (3) study of concentration and desiccation The optimum temperature was 60℃. (4) Study of Quality Standards Visible spectrophotometry and HPLC method was established for the determination,the purity of flavonoids extract was 82.38%,The average recovery rate was 97.4%,while RSD was 1.23%.Conclusion:Process was reasonable,reliable,accurate and provided a scientific and rational,practical basis for the development of Chinese herbal medicine study. |
