The preparation of pharmacodynamic components of Fructus Bruceae and Chrysomya megacephala(Fabricius)(Diptera:Calliphoridae) larvae larvae fats which have higher anti-tumor activity by enzymolysis was studied.The PB(Plackett-Burman) experiment was used to compare the significance of factors and responses surface methodology(RSM) experiment was used to optimize the factors.And the optimum enzymolysis conditions of Fructus Bruceae was adding PBS(0.01mol/L, pH 8.0) 5mL/g,petroleum ether 1mL/g and lipase 12mg/g,keeping the temperature at 38.8℃for 7h.The highest hydrolysis rate was 63.42%.The optimum enzymolysis conditions of Chrysomya Megacephala Larvae was hydrolyzing at 39.5℃for 16h40m by Candida mycoderma lipase (10mg·g-1),the ratio of raw materials to solutions was 1:7.65;the ratio of PBS(ph 8.0) to petroleum ether was 3:2.The hydrolysis rate of optimum enzymolysis process was 95.36%.The inhibitory effects of larvae oil and its enzymatic product on the tumors,of.S180 mice were tested.The anti-tumor activity of enzymatic product was 46.0%.It was much higher than that of larvae oil and similar to that of diamminedichloroplatinum(DDP).And there was a significant dose-effect relationship between anti-tumor activity and the enzymatic product.The preparation of Chrysomya megacephala larvae unsaponifiable matter was also studied.The optimum process was alkali treating for 16.5min at 56.8℃with 5%over dose alkali and petroleum ether 10mL/g. KCl solution was adding 42mL/g to break emulsion.The acid value and extraction percentage of unsaponifiable matter were 70.21mgKOH/g and 58.05%.The method of HPLC analysis was decided.The assay was conducted on Agilent XDB C18(4.6mm×150mm,5μm)column with methanol-water-acetic acid as mobile phase grade washing.The detection wavelength was270nm and pumped at 1.0mL per min.
