Studies On The DNA Damage Of Mouse Lymphocytes Induced By Herbs For Tranquilizing The Mind And Herbs For Inducing Resuscitation

In order to provide the information on the genotoxicity of the traditional Chinese medicine and guide proper clinical application,the DNA damage of mouse peripheral blood lymphocytes induced by 9 kinds of herbs for tranquilizing the mind and 7 kinds of herbs for inducing resuscitation,and the change of the DNA damage of clinical commonly used drug pair of herbs for tranquilizing the mind and herbs for inducing resuscitation were studied by single cell gel electrophoresis(SCGE).Moreover,the protective effect of the drug pair of Cinnabaris and Rhizoma Coptidis on the DNA damage of mourse cerebral cortex cells induced by Cinnabaris were also observed by SCGE in the present studies.In the study of the DNA damage induced by single herbs for tranquilizing the mind and herbs for inducing resuscitation,1/3×clinical dose,clinical dose and 3×clinical dose were used in the experiments by oral administration for 1d,3d,and 7d,respectively.The results showed that Semen Ziziphi Spinosae,Semen Platycladi,Polygala tenuifolia Willd,Caulis polygoni multiflori during these 9 kinks of herbs for tranquilizing the mind and Styrax during these 7 kinds of herbs for inducing resuscitation had no influence on DNA of mouse peripheral blood lymphocyte.However Cinnabaris,Magnetitum,Fossilia ossis mastodi,Amber,Cortex Albiziae during herbs for tranquilizing the mind and Moschus,Borneolum syntheticum,Camphor, Benzoinum,Venenum Bufonis,Rhizoma Acori Talarinowiit during herbs for inducing resuscitation induced the DNA damage of mouse peripheral blood lymphocyte with different extent.These result suggested that the DNA damage effect of animal medicinal materials and mineral medicine meterials were more serious than that of plant medicine meterials.In the following study,the reduction of DNA damage could be found in the drug pair of Cinnabaris - Rhizoma Coptidis,Cinnabaris - Magnetitum,Semen Ziziphi Spinosae - Radix Angelicae Sinensis,Moschus - Styrax however,the DNA damage effect of the drug pair of Moschus - Borneolum syntheticum were more serious than that of single medicine.Semen Ziziphi Spinosae - Polygala tenuifolia Willd,Radix Curcumae - Borneolum syntheticum did not influence the DNA damage significantly.These results indicated that the DNA damage induced by single medicine was reduced generally after the medicine was paired with other herb medicine.For all that,it should be pay more close attention that a few of drug pairs increased DNA damage compared with single medicine.In the study of the protective effect of Cinnabaris-Rhizoma Coptidis on DNA damage,the results showed that Cinnabaris could induce significantly DNA damage in mouse cerebral cortex cell which has agreement with the results in mouse lymphocytes.While the drug pair of Cinnabaris & Rhizoma Coptidis reduced greatly the DNA damage and promoted quickly the damaged DNA repairment compared with the Cinnabaris.However,the further mechanism should be study in the future.All the results demonstrated that most of herbs for tranquilizing the mind and herbs for inducing resuscitation with large dose and long term administration induced DNA damage in mouse peripheral blood lymphocytes in different degree.And the genotoxicity could be improved after most of herbs for tranquilizing the mind and herbs for inducing resuscitation paired with other herb.This study proved experimental basis for the clinical reasonable and safe application of herbs for tranquilizing the mind and herbs for inducing resuscitation...
