Traditional Chinese medicine has a long history for cancer therapy. It is drawn more and more attention because of its multi-component, multi-target, little side effects. Liriope muscari(Decne.) Baily can nourishing the Yin, promoting glandalar, miostening lung, clearing heart. This article focuses on the different extracts from Liriope muscari(Decne.) Baily, trying to find more effective anti-tumor portions.Liriope muscari(Decne.) Baily was extracted with 70% ethanol. After evaporated in vacuo, the extraction was extracted by ether, ethyl acetate,water-saturated n-butanol to get ether portion, ethyl acetate portion, n-butanol portion and water-soluble portion. Double-distilled water were added to the residues, then refluxed and evaporated in vacuo. Added alcohol to the extraction, then got 80%alcohol portion and polysaccharide portion. The result of CCK-8 experiments indicate that ethyl acetate portion has anti-tumor activity to MCF-7 cells. We explored the antitumor effect of ethyl acetate portion on MCF-7 through flurescence staining, mitochondrial membrane potential detection, Agarose gel electrophoresis, cycle detetion, Western Blot. The results indicate that ethyl acetate portion can change the morphology of MCF-7 cells significantly, induce apoptosis and G0/G1 phase cell cycle arrest and reduce mitochondrial membrane potential. Our study found that ethyl acetate portion had little effect on the expression of cyclin D1 but significantly decreased the expression of CDK4/6 and phosphorylation of Rb protein, leading to G0/G1 cell cycle arrest. In a word, our study found that ethyl acetate portion extracted from Liriope muscari(Decne.) Baily inhibited cell proliferation through promoting apoptosis and inducing G0/G1 cell cycle arrest.
