Liriope muscari (Decne.) Bailey belongs to Liriope genus of Liliaceae. It is a perennial evergreen herb and mainly distributes in Fujian province. The root has medicinal value, and it is often used to the cover ground in landscaping as the annual green. Through the ISSR molecular markers, this study analyses the genetic diversity of 31 germplasm resources of Liriope muscari (Decne.) Bailey within Fujian Province. The main research results are as follows:1. Through the monothetic experimental design, optimized each factor which influences the reactions of ISSR-PCR and established the Liriope muscari (Decne.) Bailey technology system.2. Screened out 13 polymorphic primers by using the established ISSR system of Liriope muscari (Decne.) Bailey. 13 ISSR primers producted to 162 polymorphisms from 167 sites with the 97.01% proportion. The highest percentage of polymorphisms in QuanzhouMeishan was 49.38%, and the lowest percentage in QuanzhouXiaolin and QuanzhouHutian was 38.27% polymorphisms.3. Liriope muscari (Decne.) Bailey was respectively on the research and on the analysis of genetic diversity of ISSR by POP-GENE32 software. The number of allels of the 31 germplasm resources of Liriope muscari (Decne.) Bailey was 1.9820, the efficient alleles was 1.4351, the Nei's gene diversity was 0.2774, and the Shannon's information index was 0.4358. The genetic distance of the 31 germplasm resources of Liriope muscari (Decne.) Bailey were between 0.0219 to 0.8045 and the average was 0.3105. The genetic identity were between 0.4473 to 0.9783 and the average was 0.7552. The total gene diversity was 0.3422, and the coefficient of gene differentiation was 0.6853. These showed 68.53% variation of the population genetic variation existed among populations, the genetic variation within populations was 31.47%, and the genetic variation in Gene flow among populations was 0.2296.4. Made clustering analysis of the provenances of Liriope muscari (Decne.) Bailey by the software NTSYS. The 31 provenances were divided into three kinds at 0.58 genetic similarity coefficient. The first class is YongchunShigu, YongchunDapu and Yongtai, the second class is TonganTingxi and TonganHualian, and the third class is the remaining provenances. 5. Established the digital fingerprint identifier on the germplasm resources of Liriope muscari (Decne.) Bailey based on ISSR molecular markers.
