Objective: Our aim is to establish the transplantable Kaposi’s sarcoma animal model for simulating the process of Kaposi’s sarcoma growth.Methods: 36 nude mice were randomly divided into A group, B group, C group, D group and 9 mice in each group. The logarithmic growth phase Kaposi’s sarcoma cells were calculated and regulated the cell concentration to 1×108 L-1、1×109 L-1、5×109 L-1、l×1010 L-1. 0.2ml different concentration of cells were implanted subcutaneous into the back of four groups, respectively. The formation time,changes in volume and color of tumor were observed. All mice were sacrificed when the tumor grows on day 36 and removed the tumor to analyze histopathologically. Immunohistochemical staining was applied to test the number of CD31、CD34、ki-67、Vimentin、AE1/AE3、S100、CD68、HMB-45、HHV-8.Results:(1) The Kaposi’s sarcoma model was made successfully with its cells implanting subcutaneous method, and the success rate was 100%. The results shown that after transplanting cells for six days, mice grow spots at the injection site, gradually formed plaques for 16 days, and gradually formed nodules for 23 days among mice of group A; group B is about day 4 nude mice were injected with part of a long spot, the14 days gradually formed plaques, the 21 th day gradually formed nodules. C group of about third days in nude mice injected site of long spots, eleventh days gradually formed plaques, 19 days gradually formed nodules. D group of for the second days in nude mice injected site of long spots, the tenth days gradually formed plaques, 17 days gradually formed nodules. The the development of four experimental groups tumor turned into three stages: spot, plaque, nodules. The tumor volume had a growing trend changed over time. For the moment, there was no correlation between the tumor volume and the cells concentration(P>0.05) and so did between the tumor weight on 36 th day and the cell concentration(P>0.05).Immunohistochemical staining showed positive for CD31、CD34. It proved that the tumor arised from vessels.Conclusion: The Metastatic Kaposi’s sarcoma model was made successfully with its cells implanting subcutaneous method and the higher the concentration of Kaposi’s sarcoma cells, the sooner of the formation of tumors.
