Objective: To detect the expression and the methylation status of ORF50of human herpesvirus8(HHV-8)in Kaposi’s sarcoma, and to explore the correlation and significance between the methylation andexpression of HHV-8ORF50gene and Kaposi’ s sarcoma.Methods: Extract genomic DNA, the expression of HHV-8ORF50gene was detected byimmunohistochemistry. The genomic DNA were modified by the EZ DNA MethylationTMKit. Themethylation status of HHV-8ORF50gene of55cases of Kaposi’s sarcoma and12cases of control groupwere detected by the methylation specific PCR(MSP). All the results were analyzed by the software ofSPSS17.0.Results:1. The positive of methylation of HHV-8ORF50promoter gene in Kaposi’s sarcoma has a highfrequency than control group(34.5%vs0), the difference has significant(p<0.05).2. The methylationstatus of HHV-8ORF50promoter gene between the Classic Kaposi’s sarcoma and AIDS-associatedKaposi’s sarcoma(37.8%vs27.8%) has no difference (p>0.05).3. The expression of HHV-8ORF50gene inKaposi’s sarcoma has a high frequency than control group(96.4%vs16.7%), the difference hassignificant(p<0.05).4.Tthe expression of HHV-8ORF50gene between the Classic Kaposi’s sarcoma andAIDS-associated Kaposi’s sarcoma(96.4%vs100%) has no difference (p>0.05).Conclusions:1. The demethylation and expression of HHV-8ORF50primer gene may play an importantrole in Kaposi’s sarcoma.2. HIV infection had no significant effect to HHV-8ORF50gene methylationand expression.
