Objective:The aim of the study was to compare the difference of CEUS quantitative parameters and immunohistochemical markers between the intervention-drug group and the control group, and analyze the correlation between CEUS quantitative parameters and immunohistochemical indicators, then discuss the evaluation of CEUS in the assessment of antiangiogenesis treatment in gallbladder carcinoma.Methods:Subcutaneous gallbladder carcinoma model was established. 12 tumor mass of 6 mice were selected as intervention-drug group randomly, these mice were treated with Endostar(10mg/kg/d) by intraperitoneal injection over 14 days. 20 tumor mass of 10 control mice were without any treatment. After two weeks, contrast-enhanced ultrasound was performed and image was analyzed by the software of TIC-Analysis to determine CEUS quantitative parameters(AT、TTP、PI、AUC1、AUC2). Immediately after CEUS, mice were euthanized and tumour mass removed for immunohistochemistry examination and immunohistochemical markers was measured.Results:(1) The comparison of CEUS quantitative parameters between the intervention-drug group and the control group: the mean values of PI in drug intervention group and control group were 10.82±5.46 and 16.75±5.82, respectively. The values of PI in intervention-drug group were lower than in control group significantly(P<0.05).(2) The comparison of immunohistochemical markers between the intervention-drug group and the control group: The mean values of MVD on drug intervention group and control group were 8.49±3.78 and 13.09±3.45, respectively. The mean values of VEGF on drug intervention group and control group were 4.26±0.45 and 4.71±0.43, respectively.The values of MVD and VEGF in intervention-drug group were lower than in control group significantly(P<0.05).(3) The correlation between CEUS quantitative parameters and immunohistochemical indicators: MVD and VEGF values were relevant to PI(r=0.855,P=0.003; r=0.683,P=0.042).Conclusion:(1) Compared with control group, the mean values of PI, MVD and VEGF were reduced in the intervention-drug group, so Endostar can inhibit the growth of gallbladder carcinoma.(2) There is positive correlation between CEUS quantitative parameters and immunohistochemical indicators, CEUS can be used to evaluate changes during the antiangiogenic treatment.
